The research background and significance:Oral squamous cell carcinoma(OSCC)is one of the highest incidence malignant tumor of the head/neak and maxillofacial region,multiple in adults,especially elderly male.It often occurres in the tongue,cheek,palate and so on.Advanced oral squamous cell carcinoma is often associated with lymph node metastasis,which easily lead to treatment failure,even death.Studies confirmed that the occurrence and development of oral squamous cell carcinoma is closely related to oncogenes and tumor suppressor genes,which has been widespread concern in recent years.Telomeres are special DNA sequences which located at the ends of chromosomes,which is an important part of maintaining genome stability.Telomerase maintains the length and structure of telomere,telomerase activity significantly upregulated will lead the cells? aging and death confusion,being the cancer cells.Pinx1(Pin2/TRF1 interacting protein X1)is a endogenous telomerase inhibitor,localizing at human chromosome 8p23,consisting of 7 exons,Pinx1 can reduce telomerase activity,shorten telomere length and induce apoptosis of tumor cells.Studaies found the expression of Pinx1 in the normal human tissue cells was high,but in tumor tissues the expression was less,even not,these results suggest that Pinx1 is related to the occurrence and development of cancer,so Pinx1 is expected to be a molecular marker for the diagnosis and treatment of cancer and an effective protein for molecular targeted therapy.At present,the expression of Pinx1 in oral squamous cell carcinoma has not been reported at home and abroad.Objective:By means of investigating the expression and significance of Pinx1 in different differentiation degree oral squamous cell carcinoma and normal oral mucosa,investigate the relationship between the expression level of Pinx1 and the differentiation degree and lymph node metastasis of squamous cell carcinoma,and analyze the clinical significance of the gene in oral squamous cell carcinoma from the gene and molecular level.Methods:In this study we use Immunohistochemical staining to dye and analy60 cases of oral squamous cell carcinoma tissues(High,medium and low differentiated squamous cell carcinoma 20 cases each)and 20 cases of normal oral mucosa,to detect the expression of Pinx1 in different tissues.SPSS19.0 software was used for statistical analysis,the c 2 test was used to analyze the expression level of Pinx1 in each group,P<0.05 was used as a criterion for statistical differences,to explore the correlation between the expression level of Pinx1 and the differentiation of oral squamous cell carcinoma and lymph node metastasis.Results:1.Pinx1 protein was highly expressed in all of the normal oral mucosa,the expression level of oral squamous cell carcinoma was lower than that of normal oral mucosa(P < 0.05).2.With the malignant degree of oral squamous cell carcinoma increase,Pin X1 expression level showed a decreasing trend(P < 0.05).3.The expression level of Pinx1 in oral squamous cell carcinoma tissues with lymph node metastasis was significantly lower than that in oral squamous cell carcinoma without lymph node metastasis(P < 0.05).4.The expression of Pinx1 was no significant correlation with patients? age and gender(P>0.05).Conclusion:The results of the study showed that the expression of Pinx1 was not related to patients? age and sex.The expression of Pinx1 in oral squamous cell carcinoma was significantly lower than that in normal oral mucosa.With the malignant degree of oral squamous cell carcinoma increase and lymph node metastasis,Pinx1 expression level showed a decreasing trend.These results suggest that the expression of Pinx1 is closely related to the occurrence and development of oral squamous cell carcinoma,Pinx1 is expected to become the molecular marker for diagnosing the oral squamous cell carcinoma and distinguish the malignant degree,also it may be an important target gene for gene therapy.
