Objective:To study the possible mechanisms of cluster needling of sc-alp acupuncture combined with rehabilitation on cognitive funct-ion impairment after chronic cerebral hypoperfusion by observin-g cluster needling of scalp acupuncture combined with rehabilita-tion on cognitive function?cerebral white matter?expression of corpus callosum oligodendrocytes(OLGs)in rats after chronic ce-rebral hypoperfusion.Methods:30 healthy male Wistar rats were randomly divided into sha-m operation group(Sha),model group(Mod),rehabilita tion treatme-nt group(Reh),acupuncture treatment group(Acu)and acupuncture combined with rehabilitation treatment group(A+R),six rats in ea-ch group.In addition to sham operation group,other groups were used permanent bilateral common carotid arteries ligation metho-d(2-VO)to make chronic cerebral hypoperfusion model;sham ope-ration group only separate bilateral common carotid artery(CC-A),neither ligation and not cut.After 4 weeks of cluster needlin-g of scalp acupuncture combined with rehabilitation,Morris wate-r maze test cognitive function;HE staining observe the morpho-logical changes in cerebral white matter;immunohistochemical method detect expression of positive number of corpus callosum oligodendrocytes(OLGs)in rats.Results:1.The ability of learning and memory changes:after making chronic cerebral hypoperfusion model,Morris water maze evaluate rats escape incubation period extended,the original search numbe-r less,the difference was statistically significant compared with Sha(P<0.05).After 4 weeks inter vent ion,Reh,Acu and A+R three groups escape the incubation period shorten,the original search number increased,the difference was statistically significant com-pared with Mod group(P<0.05),and A+R group Morris water ma-ze evaluating result is better than that of Reh,Acu group.2.HE staining observed:Sha cerebral white matter has no ob-vious pathology change;Mod cerebral white matter appear differ-ent degree of pathology changes:subcortical?corpus callosum,ve-ntricle narrator Cerebral white matter loose,fiber array messy,co-rpus callosum visible cavitation,sub cortical appear larger soften-ing oven,etc.Reh,Acu and A+R cerebral white matter patholog-y change is lighter,nerve fibers arranged closely,corpus callosu-m cavitation area is small,it has clear difference compared with the Mod,and A+R corpus callosum cavitation area is significantl-y reduced,closer alignment rules.3.The immunohistochemical results:cluster needling of scalp acupuncture combined with rehabilitation can increase the expres-sion of OLGs after chronic cerebral hypoperfusion in rats corp-us callosum,Reh,Acu and A+R compared with Mod,the difference was statistically significant(P<0.05).A+R expression of OLGs ab-ove Reh and Acu,the difference was statistically significant(P<0.05).Conclusion:1.Cluster needling of scalp acupuncture Combined with reha-bilitation can improve cognitive function due to chronic cerebr-al hypoperfusion in rats,and treatment effect is superior to singl-e acupuncture and single rehabilitation.2.Cluster needling of scalp acupuncture combined with reha-bilitation has certain repair effect to cerebral white matter lesi-ons in cognitive impairment rats after chronic cerebral hypoper-fusion.3.Cluster needling of scalp acupuncture combined with reha-bilitation can increase the expression of OLGs of cerebral white matter due to chronic cerebral hypoperfusionr.
