| ObjectiveBy observing the effects of Acupuncture-rehabilitation therapy on cognitive dysfunction and p-AKT and p-CREB expression in corpus callosum area of Chronic cerebral hypoperfusion rats,the efficacy of Acupuncture-rehabilitation therapy on cognitive impairment in rats with Chronic cerebral hypoperfusion(CCH)was verified,and the mechanism of acupuncture on cognitive impairment caused by Chronic cerebral hypoperfusion was discussed.This provides a theoretical basis for the treatment of cognitive impairment caused by chronic cerebral hypoperfusion with acupunctureMethodsThirty healthy male clean grade Wistar rats were divided into five groups according to a random number table,with six in each group.They were:sham operation group,model group,rehabilitation group,acupuncture group and acupuncture-rehabilitation group.Two Vessel Occlusion Groups(2-vo)were used to prepare a chronic cerebral hypoperfusion model.Morris water maze was used to detect the ability to acquire learning and memory before and after the model preparation and after treatment.Western Blot was used to detect the expression levels of p-AKT and p-CREB proteins in the corpus callosum after treatmentResults1.Changes in learning and memory acquisition ability:Morris water maze test showed that 4 weeks after the preparation of 2-vo model,the avoidance latency period of Chronic cerebral hypoperfusion rats was shortened,and the number of round platform searches was statistically significant(P<0.05).After 4 weeks of intervention treatment testing groups treatment group(acupuncture group,rehabilitation,acupuncture-rehabilitation group)rat escape latent period compared with model group,the round platform search times compared with the model group increased,statistically significant difference(P<0.05),and the acupuncture-rehabilitation,relatively simple set of the curative effect of acupuncture and rehabilitation group significantly(P<0.05)2.P-AKT and p-CREB protein expression in the corpus callosum region:compared with the sham operation group,p-AKT and p-CREB protein expression in the corpus callosum tissues in the model group decreased in proportion to the total protein expression,with statistically significant differences(P<0.05).After treatment,the difference between the treatment groups was statistically significant(P<0.05).Among them,the change of protein expression in acupuncture-rehabilitation group was significant,and the difference was statistically significant(P<0.05)Conclusion1.Acupuncture-rehabilitation therapy can shorten the escape incubation period of rats with chronic cerebral hypoperfusion,increase the number of crossing platforms,and improve the cognitive function of rats with chronic cerebral hypoperfusion2.Acupuncture may promote the phosphorylation of downstream AKT and CREB molecules in the PI3K/AKT/CREB signaling pathway,up-regulate the expression of p-AKT and p-CREB proteins,inhibit cell apoptosis,and improve the cognitive function caused by chronic cerebral hypoperfusion... |
