| Objective:The purpose of this experiment is to observe the effect of acupuncture and rehabilitation therapy on cognitive dysfunction and XIAP and Smac protein in chronic hypoperfusion rats at callosum,and to evaluate the therapeutic effect of acupuncture and rehabilitation therapy on cognitive dysfunction in chronic hypoperfusion rats.To compare the differences among acupuncture and rehabilitation therapy and simple acupuncture treatment or rehabilitation training,to highlight the advantages of acupuncture and rehabilitation therapy in the treatment of cognitive dysfunction,and to provide the relevant experimental basis for the treatment of cognitive dysfunction caused by chronic hypoperfusion.Methods:30 healthy male Wistar rats were randomly divided into sham operated group,model group,rehabilitation group,acupuncture group,acupuncture and rehabilitation group,each group contained 6 rats.Except the sham operated group,the chronic hypoperfusion model was established by the bilateral common carotid artery permanent ligation(2-VO)in the other groups,and the sham operated group only was separated the bilateral common carotid artery(Common carotid artery),neither ligation nor cut short.After 4 weeks of the treatment for four weeks according to the experimental plan about the rats in each group,the Morris water maze was used to test the cognitive function of the rats in each group.Western blot method was used to detect the levels of XIAP and Smac protein in the callosum of rats.Results:1.The change of learning and memory ability:four weeks later after the establishment about the model of chronic hypoperfusion rats,the Morris water maze test results show the prolongation of escape latency and the decrease of the frequency of the original platform.The difference between the sham-operated group and operation group was statistically significant(P <0.05).After four weeks of treatment,the results of the escape latency of the rats among the acupuncture group,the rehabilitation group,the acupuncture and rehabilitation group were shortened,and the times of the search for the original platform were increased.The difference was statistically significant compared with the model group(P < 0.05),and the test scores of the acupuncture and rehabilitation group were better than those in the simple acupuncture group and the rehabilitation group.2.Observation of corpus callosm by HE staining:there is no pathological changes were found in the corpus callosum of rats in sham operation.Pathological changes were observed in model group rats.Less pathological changes among acupuncture group,rehabilitation group and acupuncture and rehabilitation group.And the difference is obvious when compared with the model group.The pathological changes about the acupuncture and rehabilitation group were minimum and there are significant difference among acupuncture group and rehabilitation group.3.Western blot method test results: Compared with the sham operation group,the levels of Smac protein were increased and the levels of XIAP protein were decreased at the callosum of the model group,and the difference was statistically significant(P<0.05).The difference among acupuncture group,rehabilitation group and acupuncture and rehabilitation group was statistically significant compared with model group(P<0.05).The change of levels about XIAP and Smac protein in Acupuncture and rehabilitation group was more significant than that in acupuncture group and rehabilitation group(P < 0.05).Conclusion:1.Acupuncture-rehabilitation therapy significantly improved the pathological changes of corpus callosum in rats with chronic hypoperfusion.2.Acupuncture and rehabilitation therapy reduce the levels of Smac protein and increase the levels of XIAP protein at Callosum Chronic about chronic hypoperfusion rats.3.Acupuncture and rehabilitation therapy can improve cognitive function in chronic hypoperfusion rats,and the effect is better than acupuncture treatment or rehabilitation treatment. |
