| OBJECTIVE:To explore the expression of BRAF V600 E and Ki-67 in papillary thyroid carcinomas and clinical significances.METHODS:Collected the clinical data of 162 cases of thyroid papillary carcinoma reated in No.1 Hospital of Jilin University from October 2015 to September 2016,the inclusion criteria were the completion of surgery by the same surgeon and the diagnosis made by the same pathologist.Application of real-time fluorescence quantitative polymerase chain reaction(PCR): analysis of 162 cases with postoperative pathology confirmed as primary papillary thyroid carcinomas expression of BRAF V600 E gene mutation,and analysis of the correlation between its expression and clinical pathological characteristics.Application of immunohistochemical method: analysis of the 162 cases expression of Ki-67 protein and analysis of the correlation between its expression and clinical pathological characteristics.RESULTS:The results of Quantitative Real-time PCR shows that the positive rate of BRAF gene was 84.6%(137/162)in study.We analyzed BRAF V600 E gene mutation expression and relationship between clinical pathological variables,including gender,age,tumor invasion,number of lesions,tumor diameter,vessel or nerve infiltration,lymph node metastasis,presence of nodular goiter or Hashimoto thyroiditis,and clinical stage.Statistical analysis results show that except the tumor diameter between BRAF V600 E gene mutant negative group and positive group(χ2=8.550,P=0.003,P<0.05),which the higher the tumor diameter,the greater the mutated BRAF gene mutation positive rate,and there’s no significant difference in the other clinical and pathological factors(P>0.05).The results of immunohistochemical: the average expression level of Ki-67 was3.64%.In this study,we analyzed Ki-67 labeling index and relationship between the same clinical pathological variables.Statistical analysis results show that except tumor diameter,lymph node metastasis,nerve or vascular infiltrations and with hashimoto’s thyroiditis,there’s no significant difference in the other clinical and pathological factors(P>0.05).In patients Ki-67 labeling index,with lymph node metastasis,nerveor or vascular infiltrations and with hashimoto’s thyroiditiscapsule was significantly higher than those without those.There was a positive correlation between Ki-67 labeling index and tumor diameter(P <0.05),the correlation coefficient r =0.189.CONCLUSIONS:1,BRAF V600 E gene mutation is highly expressed in papillary thyroid carcinomas tissue,and BRAF V600 E gene mutation is only related to tumor diameter;2,The expression level of Ki-67 protein in papillary thyroid carcinomas was low,and its expression level was related to tumor diameter,lymph node metastasis,nerve or vascular infiltrations and hashimoto’s thyroiditiscapsule.It was an important factor influencing the infiltration and metastasis of tumor cells. |
