Objective To examine the relationship between BRAFV600E gene mutation and clinical biological characteristics in papillary thyroid carcinoma.Methods Detailed clinicopathological data of patients with thyroid papillary carcinoma who received surgical treatment in the affiliated hospital of north China university of science and technology on January 1,2011 and December 31,2015 were collected,and cancer tissue samples were collected.DNA of cancer tissues was extracted and the target gene was amplified.The BRAF mutation was detected by Sanger sequencing method,and the relationship between BRAFV600E mutation and clinical biological characteristics was analyzed.Results A total of 90 patients with thyroid papillary carcinoma who underwent surgical treatment between January 1,2011 and December 31,2015 were collected.Among them,53 patients showed positive mutation of BRAFV600E gene and 37 patients showed negative mutation of BRAFV600E gene.The positive rates of patients<45 years old and?45 years old were 32%?8/25?and 69.2%?45/65?,respectively.BRAFV600E was significantly correlated with age?P=0.001?.The positive rates in the group with and without lymphocytic thyroiditis were 76.3%?29/38?and 46.2%?24/52?,respectively.The BRAFV600E mutation was significantly correlated with the presence of lymphadenitis?P=0.004?.The positive rates were 81.0%?17/21?and 52.2%?36/69?,respectively,in the group with and without extraphyseal invasion.BRAFV600E mutation was significantly correlated with the presence of extraphyseal invasion?P=0.036?.The positive rate was69.6%?32/36?and 47.7%?21/44?in the group with and without lymph node metastasis,and the BRAFV600E mutation was significantly correlated with the presence of lymph node metastasis?P=0.035?.The positive mutation rate of BRAFV600E gene in female and male patients was 59.4%?41/69?and 57.1%?12/21?.There was no correlation between BRAFV600E mutation and gender?P=0.853?.Cancer diameter?2?2cm<d?4cm?>4cm positive rate was 57.8%?37/64?,57.1%?8/14?,66.7%?8/12?.There was no correlation between BRAFV600E mutation and tumor diameter?P=0.837?.The positive rates of single-focus group and multi-focus group were 54.5%?36/66?and 62.5%?15/24?,respectively.There was no correlation between BRAFV600E mutation and the number of lesions?P=0.501?.The positive rates in the group with and without nodular goiter were55.6%?15/27?and 60.3%?38/63?,respectively.There was no significant correlation between BRAFV600E mutation and nodular goiter?P=0.674?.The positive rates of postoperative recurrence and distant metastasis were 88.9%?7/8?and 55.6%?46/82?,respectively,and there was no significant correlation between BRAFV600E mutation and distant metastasis and recurrence?P=0.178?.Conclusions 1 BRAFV600E mutation was significantly associated with age,extramural invasion,lymphocytic thyroiditis,and cervical lymph node metastasis.2 No significant correlation was observed between BRAFV600E mutation and gender,tumor diameter,number of lesions,nodular goiter,distant metastasis or recurrence.Figure3;Table3;Reference 67.
