Puropose: This study uses the means of integration the high glucose and high lipid diet and injecting streptozotocin(STZ)to build the type ? diabetes mice model.I also observe how aerobics and metformin effecting the type ? diabetes mice fasting blood-glucose(FBG)?fasting insulin(FINS)?plasma endothelin-1(ET-1)?nitric oxide(NO)?Vascular endothelial growth factor of the skeletal muscle(VEGF),compare the difference between the diabetes group and the normal control,do the correlation analysis,and study whether the Aerobic exercise combined with the metformin can increase the effect of treating diabetes.Methods: Preparing 50 6 weeks old male SD rats,randomly selecting 10 rats as the normal control(NC,n=10),and feeding them with the standard rat feed.Feeding the other 40 rats with the high glucose and high lipid dietfor 6 weeks,and injecting the35mg/kg STZ liquor into the Abdomen according to the weight of rats,building the type ? diabetes mice model.Then randomly selecting 10 mice from then 40 as the diabetes control.The other 30 will be divided into 3 groups evenly,as diabetes aerobic exercise group(DCE,n=10)?diabetes metformin group(DCM,n=10)?diabetes aerobics and metformin group(DCEM,n=10).Giving the mice in DCE and DCEM 6 weeks' swimming training,and measuring the mice's fasting blood-glucose(FBG)?fasting insulin(FINS)?serum endotheliolysin-1 in skeletal muscle(ET-1)?nitric oxide(NO)?Vascular endothelial growth factor(VEGF).Results:(1)The change of FBG?FINS and insulin resistance index:comparing the mice in DC with the mice in NC,FBG?FINS?insulin resistance index all significantly increased(P<0.01);by the method of two-factor covariance analysis,after the 6weeks' aerobic swimming exercises intervention,FBG of T2 D mice decreased significantly(P<0.01),FINS and insulin resistance index decreased significantly(P<0.05);by 6 weeks' metformin gavage,FBG?FINS and insulin resistance index of T2 D mice decreased significantly(P<0.01);and by the intervention of aerobics and metformin,a significant interaction has been done to decreased the FBG?FINS and insulin resistance index of T2 D mice(P<0.01).(2)The change of plasma ET-1?serum NO:comparing with NC,the ET-1of DC increased significantly,NO decreased significantly(P<0.01);by the method of two-factor covariance analysis,after the 6 weeks' aerobic swimming exercises intervention,the ET-1of DCE mice decreased significantly(P<0.05),NO increased ?ET/NO decreased,but the difference is not obvious;after the 6 weeks' metformin gavage,the ET-1?ET/NO of mice in DCM decreased significantly(P<0.01,P<0.05),NO increased significantly(P<0.05);and by the intervention of aerobics and metformin,DCEM has some significant interactions for plasma ET-1?serum NO and ET/NO(P<0.01,P<0.05,P<0.01).(3)VEGF protein expression:comparing with NC,skeletal muscle of the mice in DC had the VEGF Significant expression(P<0.01),And it has statistical significance?by the method of two-factor covariance analysis,after the 6 weeks' aerobic swimming exercises intervention,skeletal muscle of the mice in DCE has the significant decrease of VEGF protein expression's average gray value(P<0.05);after the 6weeks' metformin gavage,skeletal muscle of the mice in DCE has the significant decrease of VEGF protein expression's average gray value(P<0.05);and by the intervention of aerobics and metformin,it has the significant interaction for the T2 D mice in DCEM(P<0.01).Conclusions:(1)The increasing of FBG?FINS increased means the success of ?type 2 diabetes mice model building.(2)For the type 2 diabetes mice,the increasing of ET-1,decreasing of NO,and protein overexpression of VEGF,means the dysfunction of Vascular endothelium.(3)By 6 weeks' intervention of aerobic swimming exercises and metformin,we can effectively reduce blood sugar of type 2 diabetes mice,Improve glucose metabolism of type 2 diabetes mice.Aerobics combined with the metformin can effectively increase NO,decrease the ET-1and VEGF protein expression levels in skeletal muscle.
