Objective:To analyze and discuss the expression difference of long non-coding RNA(lncRNA)of clear cell renal cell carcinoma(ccRCC)and non-tumor(NT)samples,and possible molecular mechanisms,screen out lncRNA genes,and preliminarily verify and establish a reliable lncRNA expression profiles of ccRCC.Methods:LncRNAs and mRNAs expression profiles in ccRCC samples compared with non-tumor(NT)samples were studied by high-throughput microarray.The interesting candidate lncRNAs were verified by real-time quantitative PCR.In addition,Gene Ontology(GO)and pathway analyses for differentially expressed mRNAs were performed using the standard enrichment computational method and a co-expression network of these coding-noncoding genes was constructed in our work.The Pearson correlation method was used to analyze the significant relation between the lncRNA level and ccRCC transfer.The Kaplan-Meier survival analysis was used for the survival temporal correlation.Results:Among the 78,243 lncRNA transcripts examined,we found 3,862 lncRNAs were differentially expressed(p < 0.05,2.0-fold change)in ccRCC samples relative to their matched counterparts,with 1,619 upregulated,while 2,243 were downregulated.Top 6 differently expressedlncRNA selected for further validation were aS follows.The upregulated lncRNAs were LINC00944,AC011196.3,SNHG12;The downregulated lncRNAs were.The expression levels of 6 lncRNAs examined by qRT-PCR were consistent with the microarray data.Through the Pearson correlation analysis,it is found that only the level of NONHSAT123350 is significantly related to the ccRCC transfer(r= 0.92,P = 0.0101),and is unrelated to a patient's sex,age and other factors.Among the patients with non-transferred ccRCC,the NONHSAT123350 high expression group accounts for 26% while the low expression group accounts for 74%.The result of survival analysis shows that,compared with the low expression group,the high expression group has a longer postoperative survival duration,and the postoperative median survival durations of the two groups are respectively 17.6 months(95% CI,13.6~21.3)and 26.9 months(95% CI,23.2~30.1);the difference is of statistical significance(P = 0.0015).Conclusion: There are plenty of 1ncRNAs and mRNAs with differential expressions in the ccRCC tissue and non-tumor tissue.Among the down-regulated lncRNAs,NONHSAT123350 shows a low expression in ccRCC tissue.Such a low expression is probably related to the malignant biological behaviors such as tumor invasion and metastasis.NONHSAT123350 has the highest expressive abundance in renal carcinoma cell line 786-O and the lowest basic expressive abundance in Caki-1,so the two cell lines may become the key cell lines for the research on the follow-up.
