Objective:(1)According to Uighur traditional medicine theory establish a rat model of abnormal Sapra syndrome,based on this establish a model of ulcerative colitis(UC)with abnormal Sapra syndrome,and use Uyghur medicine Xipayi Kuijiean to conduct whole process intervention,study inflammation factors expression regulation in the onset and development of UC and the molecular mechanism of Kuijiean treatment effect on UC.methods:(1)According to the theory of Uyghur medicine,dry-hot food,dry-hot environment,chronic stimulation factors were used to establish abnormal Sapra syndrome rat model,based on this model establish a rat model of UC with abnormal Sapra syndrome by TNBS/ethanol method,at the same time conduct intervention with using Kui Jie'an.(2)According to the general signs of rats and histopathological changes To evaluate efficacy.(3)collect each groups of rats colon tissue specimens,applying western blot technology phosphorylation level of NF-?B and Hu R,applying the real-time fluorescent quantitative reverse transcription polymerase chain reaction(q RT-PCR)technology,detect the expression level of hn RNA and m RNA in the five inflammation factor such as IL-1??IL-1??IL-10?IL-17?TNF-? in UC with abnormal Sapra syndrome model group,normal group,whole process intervented UC with abnormal sapra syndrome group and negative control group.Results:(1)UC with abnormal Sapra syndrome model was established successfully,and its Symptoms,pathological changes were conform to the standard of model identification.(2)compared with UC negative control group,the whole process intervented UC with abnormal sapra syndrome rats all the Symptoms have improved significantly.(3)Compared with the normal group,the phosphorylation level of NF-?B and Hu R are raised,Compared with UC negative control group,the phosphorylation level of NF-?B and Hu R were decreased in the colon tissue of UC syndrome whole process intervented group.(4)Compared with the normal group,the expression level of hn RNA and m RNA of IL-1?,IL-1?,IL-10,TNF-?,m RNA of IL-17 in UC with abnormal sapra syndrome group rats colon tissue are raised.The differences were statistically significant(P < 0.05),While the expression level of IL-17 hn RNA did not change significantly,and no statistically significant difference(P > 0.05).(5)Compared with UC negative control group,the expression level of IL-10 and TNF-? hn RNA,and IL-1?,IL-1?,IL-17,TNF-? m RNA levels were decreased in the colon tissue of UC syndrome whole process intervented group,The difference was statistically significant(P < 0.05),While the expression level of IL-1?,IL-1 ?,IL-17 hn RNA and IL-10 m RNA did not change significantly,and no statistically significant difference(P > 0.05).(6)in colon tissue of UC with abnormal Sapra syndrome group rats IL-1?,IL-1?,IL-17 m RNA expression levels were significantly higher than their hn RNA expression level,and the difference was statistically significant(P< 0.05),while There was no significant difference between IL-10 and TNF-? m RNA and hn RNA expression level(P>0.05).(7)There was no significant difference between the expression level of hn RNA and m RNA of IL-1?,IL-1?,IL-10,IL-17 and TNF-? in the rats colon tissue from whole process interfered UC with abnormal abnormal Sapra syndrome group,and no statistically significant difference(P>0.05).Conclusion:(1)IL-1??IL-1??IL-10?IL-17?TNF-? Cytokines are involved in the process of UC with abnormal Sapra syndrome generation and its recovery,After intervention with Kuijiean,The immune dysfunction tends to be basically restored in colon tissue of the UC with abnormal sapra syndrome model rats.(2)Generation of UC with abnormal Sapra syndrome and treating effects of Kuijiean are associated with NF-?B and Hu R phosphorylation.espcially,associated with TNF-? gene transcriptional activity and IL-1?,IL-1?,IL-17 m RNA stability.
