Objective:To establish the rat ulcerative colitis(UC)model and the method of intervention by Uyghur Xipayi KuiJiean for observing the expression of COX-2 mRNA in colonic mucosal tissue of UC and its correlation with effect of Uyghur KuiJiean in order to exploring the role of COX-2 in the pathogenesis of UC;to construct the recombinant eukaryotic expression vector pcDNA3.1/CT-GFP-TOPO-COX-2 bearing fusion gene of rat COX-2 gene and a green fluorescent protein reporter gene(GFP gene) for transduction it into colonic tissue of rat with genegun in vivo and investigating the expression of GFP protein,so as to inquire into the sensitive point of UC and find the gene target of Uyghur Xipayi KuiJiean.Method:Ulcerative colitis(UC)rat model was established and randomly divided into the normal group,the Ulcerative colitis(UC) model pathological group,the KuiJiean intervention group,the 5-aminosalicylic acid (5-ASA)positive controlled group,normal saline(NS)negtive controlled group,five groups.The expression of COX-2 mRNA in colon tissue of five groups was examined by RT-PCR;the recombination rat eukaryotic plasmid pcDNA3.1/CT-GFP-TOPO-COX-2 was constructed by gene engineering and molecular clone technique,then transducted into colonic tissue of rat with genegun in vivo and investigating the expression of GFP protein by Western blot.Results:In comparison with the normal group and normal saline (NS)negtive controlled group,the expression of COX-2 mRNA in the Ulcerative colitis (UC)model pathological group was increased(P<0.05).In comparison with the(UC) model pathological group,the expression of COX-2 mRNA in KuiJiean group and the 5-aminosalicylic acid(5-ASA)positive controlled group were decreased(P<0.05).By transducting the recombination rat eukaryotic plasmid into colonic tissue of rat with genegun in vivo,the expression of GFP protein could be investigated by Western blot after 24h.Conclusion:COX-2 plays a role in the pathogenesis of UC,therefore,restrain-ing the expression of COX-2 may be one of cutative mechanism of Uyghur Xipayi KuiJiean.In order to study on its molecular biology function,we succeeded in constructing the recombination rat eukaryotic plasmid and transducting it into colonic tissue of rat with genegun in vivo.On the basis of all these,we could definited the gene targts of Uyghur Xipayi KuiJiean.
