Objective:To detect the expression of phosphoinositol-3 kinase regulatory subunit2(PIK3R2)protein and mRNA in the kaposi's sarcoma,analyzing the relationship and significance between expression and different clinical stages and pathological features of kaposi's sarcoma.Methods:Selecting paraffin embedding tissue of thirty kaposi's sarcoma patients,as case groups and twenty normal skin as normal group of Uyghur nationality,we use following two methods including immunohistochemical and real-time fluorescence quantitative polymerase chain reaction(qPCR)to detect PIK3R2 expression level.Then to analyze the relationship between PIK3R2 expression level and clinical phenotype such as gender,age,pathological staging,lesion area,HIV infection.Results:Immunohistochemistry revealed that the positive rate of PIK3R2 was60% in kaposi's sarcoma tissue,while positive rate of normal skin were 25%.There was statistical significance between case groups and normal group(P<0.05).In kaposi's sarcoma group,the positive expression rate of patch was 33.3%,while positive rate of plaque period and nodular stage were respectively 71.4% and 81.8%.There was statistical difference But there were nothing in gender,age,lesion area,HIV and HHV-8 infections(P>0.05).Real-time fluorescent quantitative PCR showed that expression level of PIK3R2 was higer in Kaposi's sarcoma group than normal skin group[(2.92±0.56)vs(1.13±0.83)],with statistically significant(P<0.05).In Kaposi's sarcoma group,the expression level of patch period was lower than plaque(2.83±0.54)and nodules period(3.56±0.23),with statistically significant(P<0.05).There were no statistical significance in gender,age,lesion area,HIV and HHV-8infections(P>0.05).Conclusion:The expression level of PIK3R2 in kaposi's sarcoma is higher than the normal skin tissue.The expression of PIK3R2 is low in patch period,while the one is higher than them.PIK3R2 expressions play an important role in the process of disease development,Which provide the diagnosis and treatment of kaposi's sarcoma clues and may be used as an important index of molecular diagnostics.
