Eurycoma longifolia Jack,also known as natural viagra,Malay,native ginseng,which is together with the cubilose,pewter called the 'three national treasures' in Malaysia.The whole plant of Eurycoma longifolia Jack can be used as medicine,but usually use the root.Eurycoma longifolia Jack was used as drugs and supplements has hundreds-years history in South-East Asia,and it can be used as the single herb or party of prescription.Eurycoma longifolia Jack has the activity of anticancer and antimalarial,as well as it can improve male sexual dysfunction.Eurycoma longifolia Jack mainly contains quassinoids and alkaloids,and the quassinoids are the main active ingredient,Eurycomanone(EN)is one of the most representative component.This thesis has researched the main chemical composition from Eurycoma longifolia Jack Systematically,and establishes the main active ingredient:EN's content determination method,and investigates extraction and purification process with macroporous resin.These researches were aimed to perfect the International quality standards of Eurycoma longifolia Jack extraction,and provide the scientific basis for subsequent development.The main findings of research are as follows:1.Extraction,isolation and structure identification of main chemical composition from Eurycoma longifolia Jack19 compounds were isolated by medium-voltage preparative chromatography,silica gel column chromatography,recrystallization and Sephadex gel chromatography LH-20 from Eurycoma longifolia Jack.And their chemical construction had been identified by spectral analysis.16 of them were Quassinoids,including a new compound named ELI 5.2.The remaining three compounds,one was Adenosine,one was Scopolin,and the last one was Coniferin,which was first time isolated from Eurycoma longifolia Jack.2.Determination of the EN's content in medicinal materials and extracts of Eurycoma longifolia Jack with HPLCWe used EN as the reference substance,then established the determination method of EN by high performance liquid chromatography and ultraviolet detection,and determined EN's content of 8 batches of medicinal materials and 5 batches extracts from Eurycoma longifolia Jack with the new method.EN's content from different medicinal materials had big difference,The minimum was 0.43‰ and the highest was 1.64‰.But EN's content from different extracts were about 1%.3.Study of EN's extraction process from Eurycoma longifolia JackWith the transfer rate of EN as index,we used the single factor test and orthogonal test to explore the ethanol concentration,quantity of solvent,extracting time and extracting times.Then optimized the reasonable extraction process.The optimal condition of the extracting was 20-fold water,2 times with the first 2 and 1 h for the second time.4.Study of EN's purification process of macroporous resin from Eurycoma longifolia JackMacroporous resin model was screened by dynamic and static adsorption and desorption test.With the transfer rate of EN as index,we used the single factor test and orthogonal test to get the optimal purification process.The optimal purification process of HPD100 type of macroporous resin was 70%amount of saturated adsorption sample,0.25g/mL of sample liquid concentration,3BV/h of sample flow rate,eluting using the 25%ethanol,3BV/h of elution velocity.
