Objective As a traditional Chinese medicine commonly used drugs Agastache-Tangerine Peel,which fat soluble content is very high,this fat soluble content is important pharmacological active ingredients.In this topoic,the volatile oil components of Agastache-Tangerine Peel were studied,to optimize the extraction process of volatile oil,and the volatile oil components of the drug and single herb were analyzed by GC-MS.To compare the therapeutic effect of compatible or single application of patchouli and tangerine peel on model of functionality dyspepsia.Methods(1)The method of water distilling was used to optimize the extraction process of Agastache,Tangerine Peel and Agastache-Tangerine Peel,taking the yield of volatile oil as an index to study the factors affecting the extraction of Agastache-Tangerine Peel,for example,Soak time,liquid to material ratio,extraction time and so on.(2)Use the best technological conditions to extract the essential oil from Agastache,Tangerine Peel and Agastache-Tangerine Peel,the volatile oil were determined by GC-MS under mass spectrometer conditions of 70 e V,ion source temperature at 220 ?,transmission line temperature at 270?,quadrupole rod temperature at 40 ?,The scan range is m/z 50~800,and to assist the mass spectrometry search standard library NIST 11.L to match to determine the chemical composition of volatile oil.The relative percentage content of each component was determined by peak area normalization method.(3)Wistar rats were divided into six groups randomly,ten rats in each group.Except for nomal control group,tail clamp method was used to produce functionality dyspepsia in rest groups.The treatment group were oral given corresponding drug,once a day for one weeks.The pathological changes and other related indicators were observed in FD rats.The levels of motilin(MTL),gastrine(GAS)and acetyl choline(Ach)in rat plasma were determined by enzyme-linked immuno sorbent assay(ELISA);The expression of P substance(SP)and induced nitric oxide synthase(i NOS)were determined by immunohistochemical method.Results(1)The final conditions of extraction process were as follows: solvent ratio of11:1 don't soak,extracting for 7 hours.(2)There are 28,44 and 32 chemical components from Agastache,Tangerine Peel and drug pair were identified.There are 3 ingredients added,33 ingredients disappeared.(3)Compared with model group,compatible or single application of patchouli and tangerine peel could increase the levels of MTL,GAS and Ach in the plasma;inhibit expression of i NOS and increase expression of SP in the colon.Conclusion The optimal extraction conditions of volatile oil Can be used for drug pair extraction of fat-soluble ingredients.The composition and content of volatile oil has differences significantly in Agastache before and after combination with Tangerine Peel.Compatible or single application of patchouli and tangerine peel have certain treatment function on model of functionality dyspepsia,and the compatibility of patchouli and tangerine peel had better effect.
