PrefaceFunctional dyspepsia (FD) is a common syndrome. Some of the patients with FD present symptoms suggestive of delayed gastric emptying. So gastrointestinal hypomotility is considered one of the pstho-genesis. In this study, we analysis the effect of Chinese herbs Xiang Sha Ping Wei San ( XSPWS) decoction and its component herbs on gastric emptying and small intestinal propulsion motility of big molecule chromogen Blue Dextron ( BD) 2000 in the experimental animals, and analysis the degree of role of each component herb in the compound herb, and the mutual roles of them.Materials1. Experimental animals,; 280 5 - week - old male Kunming mice.2. Chinese herbs: Atractylodes rhizome 12g, Magnoliae officina-lis 9g, Tangerine peel 9g, Radix glycyrrhizae preparta 6g, Radix aucklandiae 9g, Fructus amomi 6g. XSPWS was made of the 6 Chinese herbs, and decocted into decoction which concentration was 0. 51g/ml (the concentration of every component herb was 0. 12g/ml, 0.09g/ml, 0.09g/ml, 0.06g/ml, 0.09g/ml, 0.06g/ml).3. Maker; big molecule chromogen Blue Dextron (BD) 2000, compounded into 2% BD2000 solution with distilled water.4. Cisapride was compounded into 0.15mg/ml cisapride solutionwith distilled water.5. 751G spectropbolometer .MethodsMice were randomly allocated to control group, cisapride group, and XSPWS group, and then received distilled water, cisapride, XSPWS decoction respectively by oral everyday during a period of 2 weeks. All experimental animals fasted an overnight before study. We used big molecule chromogen Blue Dextron ( BD) 2000 as the gastrointestinal transporting marker to observe the dosage of chromogen remaining in stomach and small intestinal propulsion rate after 20 minutes giving mice chromogen markers by oral.Mice were randomly allocated to control group, XSPWS group, atractylodes rhizome group, magnoliae officinalis group, tangerine peel group, radix glycyrrhizae preparta group, radix aucklandiae group and fructus amomi group, and then received corresponding decoctions which contained certain concentration Chinese herbs. The dosage of chromogen remaining in stomach and the small intestinal propulsion rate in mice were measured after a period of 2 weeks being given drugs by oral.We used the method of orthogonal design list L16(215) to determine the degree of role of each component herb in the compound herb, and the mutual roles of them. There were 6 factors and each factor had 2 levels, level 1 standing for the concentration of herb being 0, level 2 being certain concentration of each herb. We used different decoctions based the level of main effect of the orthogonal design list L16-(215) to make 16 experiments.ResultsCompared with control group, the rates of gastric chromogen emptying in XSPWS group and cisapride group were all significantly increased and their small intestinal propulsion rates increased (P <0. 05). The gastric emptying rate and intestinal propulsion rate had no significant difference between XSPWS group and cisapride group ( P > 0.05).Except radix glycyrrhizae preparta group, the gastric emptying rates of other component herbal groups were significantly higher than that of control group ( P < 0. 01 ) . Compared with control group, the small intestinal propulsion rates of atractylodes rhizome group, magno-liae officinalis group, radix aucklandiae group, and fructus amomi groups were increased significantly (P <0. 05). The intestinal propulsion rates of tangerine peel group and radix glycyrrhizae preparta group were no significant difference with that of control group.Based on the analysis of orthogonal design, we could conclude that the gastric emptying rate of six component herbs increased successively; atractylodes rhizom, fructus amomi, radix aucklandiae and tangerine peel. Magnoliae officinalis and radix glycyrrhizae preparta could not increase the rate of gastric emptying. There were wide ranging first grade mutual roles within the six component herbs.
