Objective:To explore and detect the expression of the human telomerase reverse transcriptase(h TERT m RNA)in peripheral blood of non-small cell lung cancer(NSCLC)and benign lung lesions by real-time polymerase chain reaction(Real-time PCR),and analyze their relationship with clinicopathological parameters.Method:(1)To extract the mononuclear cells from the peripheral blood of the patients who met the inclusion criteria,and obtain RNA from the cells to reverse transcribed to c DNA.Then using real-time PCR to detect the expression of c DNA came from h TERT m RNA in peripheral blood of 114 cases of non-small cell lung cancerand 35 cases of benign lung lesions,and analyze their relationship.(2)Using agarose gel electrophoresis(AGE)to detect the positive expression r ate of the target gene products which belong to non-small cell lung cancer and benign lung lesions obtained by real-time PCR,and analyze their relationship.Results:(1)Real-time PCR showed that the expression of h TERT m RNA in peripheral blood of four clinical stages of squamous carcinoma were(0.134±0.018)×10-3,(0.185±0.044)×10-3,(0.576±0.267)×10-3 and(3.409±2.544)×10-3.Excepting clinical stage ?and ?,the expression during other groups had a certain correlation(P<0.05);(2)The expression of h TERT m RNA in peripheral blood of four clinical stages of adenocarcinoma were(0.156±0.006)×10-3,(0.220±0.052)×10-3,(0.630±0.282)×10-3and(3.230±2.245)×10-3.Excepting clinical stage ?and ?,the relationships during other groups were statistically significant difference(P<0.05);(3)In the same clinical stage,there was a non-significant association between squamous carcinoma and adenocarcinoma(P>0.05).(4)The level of h TERT m RNA in peripheral blood of benign lung lesions was(0.042±0.017)×10-3.The difference between benign lung lesions and non-small cell lung cancer was significantly(P<0.05).(5)The positive expression rate of h TERT m RNA was 64.0% in non-small cell lung cancer group,while the positive expression rates of h TERT m RNA in four stages of patients with squamous cell carcinoma were 36.4%,41.7%,76.2% and 93.3%respectively,and the positive expression rates of h TERT m RNA in four stages of adenocarcinoma were 30% and 45.6% 68.4% and 86.7% respectively.However it's only 8.6% in benign lung lesions group.There was a statistically significant difference between that two groups(P<0.05).(6)The positive expression rate of h TERT m RNA in peripheral blood of patients with non-small cell lung cancer was not correlated with age,sex and pathological type(P>0.05),which was correlated with smoking,lymph node metastasis and clinical stage(P<0.05).Conclusion:(1)The expression of h TERT m RNA in peripheral blood of patients with nonsmall cell lung cancer was significantly higher than that in benign lung disease group and had a positive correlation with the clinical stage of non-small cell lung cancer.(2)h TERT m RNA in peripheral blood may be involved in the occurrence and development process of non-small cell lung cancer.(2)h TERT m RNA could be a molecular marker of non-small cell lung cancer,early screening of high-risk groups,treatment and follow-up of patients with non-small cell lung cancer.
