Objective To Observe the expression change of Th17 cells and IL-17 in rat intestinal mucosa lamina with obstructive jaundice, analysis the effects of Thl7 cells and the expression of cytokines IL-17 on intestinal mucosa barrier.Give the exogenous leptin for intervention,to explore a new protective way of leptin in intestinal mucosa.Methods 24 SPF level male SD rats were randomly divided into normal control group,obstructive jaundice group, leptin group, Set the control group to be sham group.Give obstructive jaundice group and leptin group duct ligation.Leptin group rats receive an injection with rat recombinant leptin daily lOug/kg since the first day after surgery,Other groups of rats by injected with the same amount of normal saline as control.Three groups of rats were killed in 7 days and drew materials.HE observe the morphological changes of the intestinal mucosa structure,FCM test the Th17 cells percentage changes in Intestinal mucosa lamina propria,IHC test the expression of IL-17?ZO-1 in Intestinal mucosa.Results 1. In general, 2 rats of oJ group died respectively at postoperative 5?6th with Intestinal obstruction and abdominal cavity infection, 2 rats of leptin group died respectively at 4?6th with Biliary fistula and Bilious ascites.2. Obstructive jaundice rats compared with normal rat have higher Th17 cells percentage obviously in intestinal mucosa lamina propria, but Th17 cells percentage in rats with exogenous leptin is lower compared with obstructive jaundice rats (OJ group: 3.17±0.64,Normal group: 0.398±0.152,Leptin group: 1.925±0.6 P<0.05).3. IHC: Normal control group of rats and obstructive jaundice group can be seen with the expression of IL - 17 in the intestinal mucosa, OJ group has higher IL-17 expression, but IL-17 expression in rats with exogenous leptin is lower compared with obstructive jaundice rats (Normal group: 0.79±0.07, OJ group:1.23±0.14,Leptin group: 0.97±0.11 P<0.05).4. The IL-17 expression of obstructive jaundice rat intestinal mucosa has strong correlation with serum total bilirubin leve(r=-0.7528, P<0.01). The expression of IL-17 rise with the increase of bilirubin level.5. IHC: OJ group has lower ZO-1 expression, but ZO-1 expression in rats with exogenous leptin is higher compared with obstructive jaundice rats (Normal group: 1.33±10.11,OJ group: 0.83±0.09,Leptin group:1.07±0.10 P<0.05) .6. The expression of ZO-1 has negative correlation with IL-17 expression in rat intestinal mucosa with obstructive jaundice (r=-0.66,p<0.05).Conclusion Obstructive jaundice rat intestinal mucosa lamina propria Th 17 cell proportion and IL-17 expression increased, and be negative correlation with total bilirubin level.The expression of ZO-1 has negative correlation with IL-17 expression in rat intestinal mucosa with obstructive jaundice. Speculated that obstructive jaundice Th17 under high differentiation and high expression of IL-17 may participate in the function of intestinal mucosa damage.Give exogenous leptin treatment,rat intestinal mucosa IL-17 expression decreased,speculated that inhibit Th 17 cells differentiation of quantity and reduce the expression of cytokines IL-17 is one of the mechanism of leptin to maintain the integrity of intestinal mucosa.
