Objective:To investigate the regeneration of pulp-dentin complex which loading the stem cells of apical papilla on platelet-rich plasma scaffold in empty root canal,and implanting the root in SD rat subcutaneous tissue,The accomplishment of the proposed study will provide a theoretical basis and the foundation of research for the clinical application.Method:1.Isolation,culture and identify of the SCAP: Collect the papilla tissue from the permanent teeth which remove because of orthodontic treatment,isolate,culture and identify the SCAP.2.Root canal preparation: Remove the crown and apical of the anterior teeth,preparing the root canal to 200#,extract the antigen of the root.3.platelet-rich plasma scaffold preparation :Centrifuge the whole blood of SD rats,Mixing the plasma and platelets to get the PRP.4.loading the stem cells of apical papilla on platelet-rich plasma scaffold in empty root canal,and implanting the root in SD rat model,dividing in to 3 experimental groups(PRP group,SCAP+DMEM group and SCAP+PRP group),the empty root l as control.Killing the SD rats and collecting the root after 2 months,to observe the pulp-dentin complex regeneration by sections.Result1.Control group: the fibrous tissue grew in to the root canal,and the formation of pulp-dentin complex was not observed.2.SCAP+DMEM group: the cells were implanted in the root canal,but there was no obvious angiogenesis,A large number of blue-stain collagen fibers can be see in the selection which indicated lots of extracellular matrix were be produced.3.PRP group: there was no obvious angiogenesis in root canal too,the cells were implanted in the root canal,but there was no like pulp structure formation,we find the light red-stain matrix in the selection.4.SCAP+PRP group: there was a new tissue formation in the root canal,and we find out like pulp-dentin complex,we can see new blood vessels in the intercellular substance,a large amount of extracellular matrix and collagen fibers were produced.Conclusion:There was no obvious angiogenesis in the control group,PRP group and SCAP group.There was like pulp-dentin complex formation in the selection,a large amount of extracellular matrix and collagen fibers were produced.
