Oleanolic Acid On Androgen Alopecia Treatment Function And On Expression Effect Of VEGF And TGF-?1 In Androgenetic Alopecia Lesions Of Mice

Object:With the androgenetic alopecia mice models for the study objectives,considering the expression of VEGF and TGF-?1,and the treatment function of oleanolic acid on androgen alopecia mice and the expression of VEGF and TGF-?1 after the treatment with oleanolic acid.Then investigating the pathogenesis-related effects of VEGF and TGF-?1 in androgenetic alopecia mice and oleanolic acid on the level of their expression.Methods:Subcutaneous injection of testosterone propionate in mice induced androgenetic alopecia animal models,then they were taken into groups after successful modeling.Each group was given a different treatment according to the experimental requirements.At 8th and 12th week,for each group,mice's lesions were taken for HE staining to measure dermal thickness and the number of hair follicles,and immunohistochemistry was used to detect the expression of VEGF and TGF-?1 on the dermis around the lesions of the normal group,androgenetic alopecia group and oleanolic acid and minoxidil group.Results:At the 6th week,the number of hair follicles were significantly reduced in the skin and dermal thickness was thinner in the model group than the control group,modeling was successed.After comparing the results of the 8th and 12th week,we found that the VEGF expression level of lesions in model group was significantly lower than the normal control group at 8th week,while the TGF-?1 expression was significantly higher.And at the 12th week,the TGF-?1 expression increased further over its 8th week,the difference was statistically significant(P<0.01).In model group the number of hair follicles in the skin decreased significantly and dermal thickness was significantly thinner than the normal control group,and the number of hair follicles at 8th week were further less than its results of 12th week,and dermal thickness was further thinning,the difference was statistically significant(P<0.01).We found that the expression of VEGF was significantly higher at 8th week in oleanolic acid lesions of mice compared with the model group,but lower than the normal control group,and its expression at 12th week further increased,being roughly close to the minoxidil group,but slightly lower than the normal control group.The expression of TGF-?1 was significantly lower at 8th week in oleanolic acid lesions of mice compared with the model group,but lower than the normal control group,and its expression level at 12th week further reduced,being roughly close to the minoxidil group but slightly lower than the normal control group,the difference was statistically significant(P<0.05).The number of hair follicles in the skin of oleanolic acid group of mice were significantly higher than the model group,and dermal thickness was significantly thicker,but lower than the normal control group at the 8th week.While the number of hair follicles and dermal thickness in the skin of mice of oleanolic acid group further increased at the 12th week,being close to minoxidil group but slightly lower than the normal control group,the difference was statistically significant(P<0.01).Conclusion:The expression of VEGF of lesions in model group was significantly lower than the normal control group,while the TGF-?1 expression was significantly higher,and the number of hair follicles in the skin were significantly less than the normal control group and dermal thickness was significantly thinner.After using topical treatment with oleanolic acid,the expression of VEGF in lesions of mice at oleanolic acid group rebounded significantly,and the expression level of TGF-?1 was significantly reduced,and the number of hair follicles in the skin were significantly higher compared with the model group,the dermal thickness increased significantly.And with the appropriate duration of treatment,the expression of VEGF in mice skin of oleanolic acid group was further recovery,while the expression of TGF-?1 was further reduced.The number of hair follicle and dermal thickness further increased,being roughly close to the minoxidil group but slightly lower than the normal control group.It is consistent with the external treatment of androgenetic alopecia in histopathology and at the molecular level.Oleanolic acid treatment with androgenetic alopecia mice was effective.
