Study On The Effect Of Xuan Fei Bu Shen Yang Yin Decoction On C-kit Protein And S-100 Protein And NNOS In Rats With Slow Transit Constipation

Abstract/Summary:
Objective: The aim of this experiment was to study the level of c-kit protein and S-100 protein and nNOS of rats with slow transit constipation,to explore the mechanism and therapeutic effect of the treatment of slow transit constipation.Methods: 60 SD rats were randomly divided into blank group and model group.Rats in model group were intragastric administration by compound diphenoxylate,the rats in the blank group were intragastric administration by distilled water.After buliding the model successfully,The rats of model group were divided into five groups according to the random principle.The rats of treatment group were intragastric administration by the corresponding drugs,meanwhile the rats of control group and blank control group were intragastric administration with distilled water,After the correspondent treatment of each group,calculate the intestinal propulsive rate of all rats,observe the c-kit protein and s100 protein in colonic tissues of rats,Observe the change of nNOS in serum.Results:(1)Compared with the blank group,The intestinal propulsive rate of model group was significantly reduced(P<0.01);The number of c-kit protein in colon tissue of model group was significantly reduced(P<0.01);The number of S-100 protein in colon tissue of model control group was significantly increased(P<0.01);The concentration of nNOS in serum of the model group had no significant difference(P>0.05).(2)Compared with the model control group,the high dose of Xuan Fei Bu Shen Yang Yin decoction group and cisapride group obviously increased the intestinal propulsive rate(P<0.01),The medium dose and low dose of Xuan Fei Bu Shen Yang Yin decoction also can improve the intestinal propulsive rate,(P<0.05);The number of c-kit protein in colon tissue of rats in Si Shah Billy group and the high dose of Xuan Fei Bu Shen Yang Yin decoction group was significantly increased,(P<0.01),The number of c-kit protein in the colon tissue of the medium dose and low dose was also be increased(P<0.05);The number of S100 protein in colon tissue of rats in Si Shah Billy group and high dose group was significantly reduced(P<0.01),The medium dose and low dose of Xuan Fei Bu Shen Yang Yin decoction also can decrease the number of S-100 protein in colon tissue(P<0.05);There was no significant difference in the expression of nNOS in serum(P>0.05).Conclusion:(1).The content of c-kit protein in the colon of rats with slow transit constipation decreesed significantly;and the number of S-100 protein increased significantly,It is suggested that c-kit protein and S-100 protein may be involved in the pathogenesis of slow transit constipation,and there is no significant difference in values between model group and blank group.The relationship between slow transit constipation and nNOS needs further study.(2).Compared with the model control group,each dose of Xuan Fei Bu Shen Yang Yin decoction can improve the intestinal propulsion rate and the content of c-kit protein in colon tissue of rats with slow transit constipation,also can reduce the content of S-100 protein in colon tissue,but the effect of high dose is most remarkable;Suggest that the effect of high dose of Xuan Fei Bu Shen Yang Yin decoction is the optimal dose;After the end of treatment,there was little change in the content of nNOS in all rats,and the specific reasons need to be further studied.
