Effects Of Xuanfei Bushen Yangyin Decoction On P-AKT Protein,SYN Protein And NPY In Rats With Slow Transit Constipation

Abstract/Summary:
Objective:By analyzing the effect of Xuanfei Bushen Yangyin decoction on the changes of P-AKT protein,SYN protein and serum NPY in proximal colon of rats with slow transit constipation(STC),the mechanism of Xuanfei Bushen Yangyin decoction in treating STC was explored.Methods:Forty-eight SD rats,half female and half male,were randomly divided into blank group and model group,rats in blank group were given intragastric administration of normal saline,rats in model group were given compound difenoxylate[1]intragastrically to observe and verify the success of the model.The rats in the model group were divided into five groups according to the principle of random distribution.The rats in the blank group and the model control group were given intragastric administration of normal saline,and the other groups were treated with the corresponding drugs.After the treatment,the rats were killed and the specimens were collected.The changes of P-AKT protein,SYN protein and serum NPY in proximal colon of rats were detected.Results:(1)Compared with the blank group,the intestinal propulsive rate in the model control group was significantly lower than that in the control group(P<0.01).Compared with the model control group,the intestinal propulsive rate of rats in each dose group of Xuanfei Bushen Yangyin decoction increased significantly(P<0.05).(2)Compared with the blank group,the content of P-AKT protein in colon tissue of the model control group was significantly higher(P<0 01).Compared with the model control group,the content of P-AKT protein in colon tissue of rats in Xuanfei Bushen Yangyin decoction low dose group and middle dose Xuanfei Bushen Yangyin decoction group was significantly lower than that in model control group(P<0 01),There was no significant change in the content of P-AKT protein in colon tissue of rats in the high dose Xuanfei Bushen Yangyin decoction group,and there was no significant difference(P>0 05).(3)Compared with the blank group,the content of SYN protein in the colon tissue of the model control group was significantly lower(P<0 01).Compared with the model control group,the content of SYN protein in colon tissue of rats in each dose group of Xuanfei Bushen Yangyin decoction was significantly higher than that in the model control group(P<0.05).(4)Compared with the blank group,there was no significant difference in serum NPY between the blank control group and the control group(P>005).Compared with the model control group,there was no significant difference in serum NPY between the groups of Xuanfei Bushen Yangyin decoction and the model control group(P>0.05).Conclusion:(1)The increase of P-AKT protein and the decrease of SYN protein in colon of STC rats suggest that P-AKT protein and SYN protein may be involved in the pathogenesis of slow transit constipation.(2)The dosage group of Xuanfei Bushen Yangyin decoction(high,middle and low)could increase the intestinal propulsive rate of STC rats,and Xuanfei Bushen Yangyin decoction could decrease the P-AKT protein of colon of STC rats in low dose group and middle dose group.All groups of Xuanfei Bushen Yangyin decoction could increase SYN protein in colon of STC rats.(3)There was no significant change of P-AKT protein in colon of STC rats in the high dose group of Xuanfei Bushen Yangyin decoction,and the content of NPY in serum of the blank group and each model group had little effect,and the specific reasons needed to be further explored.
