Studies On Immune Function And Antitumor Biological Activity Of Sturgeon Chondroitin Sulfate

Sturgeon chondroitin sulfate has many important biological functions,but little is known about its immunologic function.Moreover,chondroitin sulfate has been comfirmed to play an important role in antitumor,but the action mechanism of which has not been revealed.In this study,to study the effects of sturgeon chodroitin sulfate on immune function and anti-inflammatory action,the delayed-type hypersenditivity response,the anaphylactic reaction induced by dimethylbenzene which results in ear swelling,and foot pad induced by egg albumen were measured,and paraffin section method was used,after clean grade KM mice were administered with different doses of sturgeon chondroitin sulfate for two weeks.We also study the anti-tumor effects of sturgeon chodroitin sulfate of different doses?0.03 mg/kg,0.3 mg/kg and 3 mg /kg?on H₂₂ hepatocelluar carcinoma-bearing mice.Our works provided basic materials to know the immunologic functions and anti-tumor activities of sturgeon chondroitin sulfate,and the main results as follows:1.Compared with the control group,the sturgeon chondroitin sulfate could increase the weight of thymus and spleen?P<0.05?and the amount of the perivascular leukocyte?P<0.01?,obviously enhance the delayed-type hypersensitivity response intensity?P<0.01?,improve the productive ability of specific antibody in mice?P<0.05?and reduce the degree of ear swelling induced by dimethylbenzene and swelling of foot pad induced by egg albumen?P<0.05?.2.The sturgeon chondroitin sulfate could increase the amount of the number of mast cell in immune organs of mouse,among which,administered with the dose of 600 mg/kg sturgeon chondroitin sulfate could enhance significantly the weight of the thymus?P<0.05?and increase the number of the mast cell in the thymus?P<0.01?,and administered with the dose of 60 mg/kg could enhance significantly the weight of the spleen?P<0.01?,and most obviously,administered with the dose of 6 mg/kg could increase the number of the mast cell in the spleen?P<0.01?.3.MTT assay was carried out and the absorbance values at 490 nm were measured on an enzyme-linked immunosorbent assay to observe the effects of sturgeon chodroitin sulfate on growth of hepatoma carcinoma cell in vitro culture.The results showed that sturgeon chondroitin sulfate had no significant inhibition effect on the H₂₂7402 and H₂₂7721 cells in vitro culture,but had inhibition effect on H₂₂ Hep G2 cells in vitro culture,and the inhibitory effect on tumor cells of chondroitin sulfate with the final concentration of 1000 ?g/ml and 125 ?g/ ml was well.4.After ten days ig administration of sturgeon chodroitin sulfate with different doses,the clean grade KM mice were then injected with H₂₂ tumor cell under the skin of axillary,and the weight of mice,tumor size and weight,and the inhibition ratio were observed.The results showed that the sturgeon chondroitin sulfate could increase the weight of H₂₂ hepatocelluar carcinoma-bearing mice and could inhibit tumor growth.Among which,administered with the dose of 60 mg/kg sturgeon chondroitin sulfate could enhance significantly the weight of mice?P<0.05?and could significantly inhibit the tumor volume?P<0.05?.5.To study the effects of sturgeon chodroitin sulfate on the weight of clean grade KM mice,tumor size and weight,and the inhibition ratio,the mice were injected with H₂₂ tumor cell under the skin of axillary,after 24 h,and then were administered with different doses of sturgeon chodroitin sulfate for one week.The results showed that the sturgeon chondroitin sulfate could increase the weight of H₂₂ hepatocelluar carcinoma-bearing mice and could inhibit significantly tumor growth.Among which,administered with the dose of 60 mg/kg sturgeon chondroitin sulfate could enhance significantly the weight of the mice?P<0.05?and could significantly reduce the tumor weight?P<0.05?,and administered with the dose of 600 mg/kg sturgeon chondroitin sulfate could inhibit significantly the tumor volume?P<0.05?.
