Objective To detect the expression of p-mTOR and p-P70S6K in gastric cancer, and tumor adjacent tissues, what is more, to investigate its relationship with clinic pathological index. In order to provide a new theoretical basis for the early diagnosis,targeted therapy and prognosis of gastric cancer, we further explore the mechanism of them in the occurrence and development of gastric cancer.Methods The 71 cases with gastric cancer from the Binzhou Medical University Hospital were confirmed by pathology following the surgery. Another 15 normal gastric tissues from patients underwent surgery because of other diseases, were selected as control. Expressions of p-mTOR and p-P70S6K were detected in 71 cases of gastric carcinoma and tumor adjacent tissues by RT-PCR and immunohistochemical methods. Meanwhile, the relationship between the two indicators and clinicopathological index was analyzed.Results The expression levels of p-mTOR and p-P70S6K mRNA in gastric cancer were much higher than that in other two groups, the difference were significant(P<0.05).Expression of p-mTOR was correlation with p-P70S6K.Univariate analysis results were that p-mTOR was no correlation with age, tumor size,histological grade,but related with lymph node metastasis and clinical stages (P<0.05); In immunohistochemical protein expression level, The expression of p-mTOR and p-P70S6K in gastric carcinoma were 60.6%, 64.8% respectively, and 30.1%, 36.6%in tumor adjacent, differences are all signifigant(P<0.05).Conclusion(1) p-mTOR and p-P70S6K were significantly activated in gastric carcinoma,which indicates mTOR signal pathway plays important roles in the development of gastric carcinoma.(2) p-mTOR was correlation with p-P70S6K, which indicates that these two combine action in the gastric carcinoma.(3) The expression of p-mTOR and p-P70S6K were significantly related with lymph node metastasis and the clinical stage of tumor.
