Aim:To investigate the expression of mTOR/p70S6K signaling pathway in WHO Grade Ⅱ and Ⅲ gliomas,and evaluate the association between mTOR/p70S6K signaling pathway and the prognosis of glioma patients. Method:The expression of mTOR and p70S6K were assessed by immunohistochemical staining in148cases of newly diagnosed glioma,the Correlation of both markers was tested by Spearman correlation. Collected148cases of glioma patients medical records information, and all patients were followed for, the time from87-2177days after operation, a retrospective study was carried out in the follow-up data. The Kaplan-Meier method was applied to evaluated the relationships between the expression of mTOR and p70S6K and overall survival(OS) and progression-free survival(PFS). Cox regression was used to correlate the expression of mTOR and p70S6K with OS while adjusting for clinicopathological variables. Results:There was statistically significant difference between the expression of mTOR and p70S6K in different grades of gliomas (P<0.05),a positive linear correlation between the mTOR and p70S6K expression (r=0.596. P<0.0001). Multivariate Cox regression analysis revealed that the expression of p70S6K is an independent prognostic factor for OS of glioma patients. High p70S6K expression group have shorter overall survival time (hazard ratio:3.606;95%confidence interval:1.066-12.201;P=0.039). In contrast, mTOR expression were not associated with the prognosis of patients. Conclusion:mTOR/p70S6K signaling pathway plays an important role in the process of glioma progression.The expression level of p70S6K in predicting prognosis of patients with gliomas have clinical significance, may be potential targets for gene therapy of.
