Objective: To investigate the effects of dietary Advanced glycation end products(AGEs)make on gut microbiota and insulin resistance in C57BL/6 mice.Methods dietary AGEs diet,exogenous AGEs diet and Regular diet were conducted to C57BL/6 mice for 24 weeks.Measure the FBG,weight,serum insulin,serum AGEs and LBP level of mice,and the AGEs content of diets.Use the Illumina HiSeq sequencing the V4 region of the bacteria 16 S ribosomal RNA of gut microbiota to analyze the different structure of gut microbiota in mice consume different diets.Result:(1)exogenous AGEs diet had an AGEs content about four times that of the regular diet and about two times that of Heat-treated Diet.(2)After 12-week intervention,levels of FBG in exogenous AGEs diet group and dietary AGEs diet group were higher than control group,after 24-week intervention,the levels of FBG,fasting serum insulin,HOMA-IR in MG-BSA group and dietary AGEs diet group were significant higher than control group,the result of IPGTT exhibited glucose intolerance in two intervention group,while the weight of mice in three groups were comparable.(3)Serum AGEs level of exogenous AGEs diet group and dietary AGEs diet group were significant higher than control group,while the serum LBP level of exogenous AGEs diet group was significant higher than dietary AGEs diet group and control group.(4)After 24-week intervention,the Community richness of gut microbiota in two intervention groups were less than control group.There were 12 genus were discovered as high-dimensional biomarkers for separating gut microbiota between exogenous AGEs diet group and control group,and 2 genus were discovered between dietary AGEs diet group and control group.The genus whose relative abundance were higher in exogenous AGEs diet group were most belong to Clostridiales,while the genus whose relative abundance were higher in control group were Bifidobacterium and those belong to Bacteroidales.(5)The relative abundance of Allobaculum have a negative correlation relationship with FBG and HOMA-IR,while the relative abundance of Parasutterella and Turicibacter have a positive correlation relationship with FBG and HOMA-IR.Conclusion: Excessive intake of AGEs in C57BL/6 mice induce lower community richness of gut microbiota,altered gut microbiota structure,leading to higher serum endotoxin level and promote insulin resistance.
