Research background:Pancreatic cancer(PC)is a very high degree of malignancy with an extremely poor prognosis in human beings' digestive system.With the change of people's living and eating habits,such as intaking too much high-fat and high-energy food,excessive drinking and smoking,coupled with the issues of environmental pollution and unsafe food,more and more people are suffering from pancreatic cancer,which has been ranked fifth among cancer-related deaths in China.The symptoms of pancreatic cancer in the early stage are dormant.It is too late when the patients' symptoms are abvious,because at this time there have been local invasion and distant metastasis,resulting in low rate surgical resection and unsatisfactory chemotherapy and radiotherapy.Therefore,it is very important to concern how to solve the problem of early detection,early diagnosis and early treatment of pancreatic cancer in the world including our China.With the developing of fundamental researches about tumor,a large number of non-coding RNAs are discovered,which are related to the occurrence and progression of tumor.Those non-coding RNAs,including lncRNA,mi RNA,siRNA,pi RNA,circRNA,etc,have no or only one poorly conserved readable frame and have no function of encoding proteins.Currently,more and more attentions are focus on those non-coding RNAs.Long non-coding RNA(lncRNA)is a class of non-coding RNAs with a transcript length of more than 200 nt.4%-9% of the mammalian genome sequence produce lncRNAs,whereas only 1% of the RNAs have the function of encoding proteins.Initially,these lncRNAs are not taken seriously,because they are considered to be "noises" in gene transcriptions without biological functions.At present,more and more studies have shown that lncRNAs are closely related to the occurrence and development of tumors.These lncRNAs can be involved in the proliferation,invasion and metastasis of malignant tumors by chromatin modification,genomic imprinting,transcriptional activation,post-transcriptional regulation and protein function regulation.Therefore,our research group find out high expression of lncRNA LOC100506123 in pancreatic cancer tissues through high-throughput lncRNA gene chip,and explore the correlation between lncRNA LOC100506123 and pancreatic cancer.Objectives:To explore the expression level of long non-coding RNA(lncRNA)LOC100506123 in pancreatic cancer tissues and cell lines,and to observe lncRNA LOC100506123' effect on proliferation,apoptosis and migration of pancreatic cancer cells through down-regulating the expression of LOC100506123 at the cellular level by si RNA technique,and to explore LOC100506123' potential role in the occurrence and development of pancreatic cancer.Methods:(1)The expression profile of long non-coding RNAs in pancreatic and adjacent tissues is screened by high-throughput gene chip technique and the significant different lncRNAs are presented by bioinformatics analysis.(2)The expression of lncRNA LOC100506123 in the gene chip is verified by real-time quantitative PCR(qRT-PCR),and the correlation between the expression of lncRNA LOC100506123 and the clinical survival prognosis is analyzed.(3)The expression level of LOC100506123 is detected by q RT-PCR in pancreatic cell lines(normal pancreatic duct epithelial cell line HPDE,and 4 pancreatic cancer cells AsPC-1,Bx PC-3,Panc-1,CFPAC-1).(4)The si RNAs are used to down-regulate the expression of LOC100506123 in the pancreatic cancer cell line AsPC-1 and CFPAC-1(si R-NC in the control group,siR-1 and si R-2 in the experimental group).(5)The proliferation of pancreatic cancer cells are detected by CCK8,and the migration ability of pancreatic cancer cells is detected by Transwell Chamber Test.Results:(1)The expression of lnc RNA LOC100506123 is significantly increased in pancreatic cancer tissues,and its high expression is negatively correlated with clinical survival prognosis.(2)The expression of LOC100506123 in pancreatic cancer cell lines As PC-1,Bx PC-3,Panc-1 and CFPAC-1 is significantly higher than that in normal pancreatic duct epithelial cell line HPDE.(3)After down-regulating the expression of LOC100506123 by siRNAs,the proliferation of pancreatic cancer cell lines As PC-1 and CFPAC-1 is significantly decreased.(4)At the same time,the migration ability of pancreatic cancer cells is significantly reduced.Conclusion:LncRNA LOC100506123 is highly expressed in pancreatic cancer and closely related to the survival prognosis of clinical patients.After down-regulating the expression of lncRNA LOC100506123,the proliferation and migration of pancreatic cancer cell lines are decreased.It is suggesting that lnc RNA LOC100506123 is related to the occurrence and development of pancreatic cancer,and it may become an important target for the treatment of pancreatic cancer.
