Objective:To explore the function of IL-6 on the expression of Androgen Receptor(AR),Signal Transducer and Activator of Transcription 3(STAT3)and Mitogen Activated Protein Kinase(p38MAPK)in prostate cancer cell lines PC-3 and LNCaP.According to the results,we discuss the probable function of IL-6 on the occurrence and developement of prostate cancer(PC)via the AR?STAT3 and p38MAPK.Our study will inspire us to look for new methods to treat PC.Methods:1.After treating prostate cancer cell lines LNCaP(hormone-dependent)and PC3(hormone-independent)by IL-6 in different concentrations,we detected the cells proliferation with CCK-8.2.After treating prostate cancer cell line LNCaP and PC3 by IL-6 in different concentrations,we detected the expression of AR in two cell lines with real-time fluorescent quantitative PCR(Q-PCR).3.After treating prostate cancer cell lines LNCaP and PC3 by IL-6 in different concentrations,we detected the expression of AR?STAT3 and p38MAPK in two cell lines with western bolt(WB).Results:1.The effect of IL-6 on proliferation of prostate cancer cells(LNCaP?PC3)(1)The proliferation of LNCaP was different after treating by different concentrations of IL-6(Ong/ml,lOng/ml,20ng/ml,30ng/ml,40ng/ml,50ng/ml)for 24hs and 48hs,and the difference has the statistical significance(P<0.05).The peak of proliferation was 24hs later at 20ng/ml.(2)The proliferation of PC-3 was different after treating by different concentrations of IL-6(Ong/ml,lOng/ml,20ng/ml,30ng/ml,40ng/ml,50ng/ml)for 24hs and 48hs,and the difference has the statistical significance(P<0.05).The peak of proliferation was 24hs later at 20ng/ml.(3)The proliferation of(LNCaP?PC3)was different after treating by same concentrations of IL-6(lOng/ml,20ng/ml)for 24hs,the proliferation of LNCaP was stronger than PC3,and the difference has the statistical significance(P<0.05).2.The effects of IL-6 on transcriptional level expression of AR in PC cell lines(LNCaP?PC3)The study found that the expression of AR was increased in transcription level in LNCaP and PC3 cell lines after treating with different concentrations of IL-6 for 24hs.The difference in different concentration was statistically significant(P<0.05).The more expression of AR was correlated with the higher concentration of IL-6.The expression of AR was no significant change in transcription level in LNCaP and PC3 cell lines after treating with same concentrations of IL-6 for 24hs,and the difference was not statisticaly significant(P>0.05).3.The effects of IL-6 on translation level expression of AR,STAT3 and p38MAPK in PC cell lines(LNCaP?PC3)(1)The effects of IL-6 on translation level expression of AR in PC cell lines(LNCaP,PC3):The study found that the translational expression of AR in PC cell lines after treating with different concentrations of IL-6 for 24hs was different and the difference has the statistically significance(P<0.05).The more expression of AR was correlated with the higher concentration of IL-6.The expression of AR was different in translation level in LNCaP and PC3 cell lines after treating with same concentrations of IL-6 for 24hs,the expression of AR in translation level in LNCaP was stronger than PC3,and the difference has the statistical significance(P<0.05).(2)The effects of IL-6 on translation level expression of STAT3 in PC cell lines(LNCaP,PC3):The study found that the translational expression of STAT3 in PC cell line after treating with different concentrations of IL-6 for 24hs was different and the difference has the statistically significance(P<0.05).The more expression of STAT3 was correlated with the higher concentration of IL-6.The expression of STAT3 was no significant change in translation level in LNCaP and PC3 cell lines after treating with same concentrations of IL-6 for 24hs,and the difference has statistical significance(P>0.05).(3)The effects of IL-6 on translation level expression of p38MAPK in PC cell lines(LNCaP,PC3):The study found that the translational expression of p38MAPK in PC cell line after treating with different concentrations of IL-6 for 24hs was different and the difference has the statistically significance(P<0.05).The more expression of p38MAPK was correlated with the higher concentration of IL-6.The expression of p38MAPK was different in translation level in LNCaP and PC3 cell lines after treating with same concentrations of IL-6 for 24hs,the expression of p38MAPK in translation level in PC3 was stronger than LNCaP,and the difference has the statistical significance(P<0.05).Conclusions:1.IL-6 promotes the proliferation of androgen-dependent prostate cancer cell(LNCap)and androgen-resistant prostate cancer cell(PC-3).2.The AR,STAT3 and p38MAPK were expressed in hormone-dependent cell line(LNCaP)and hormone-independent prostate cancer cell line(PC3).3.The transcriptional expression of AR in LNCaP and PC-3 cell lines was significant after treating with different concentrations of IL-6 for 24hs.The difference was statistically significant(P<0.05).The expression of AR was correlated with the higher concentration of IL-6.4.The translational expression of AR,STAT3 and p38MAPK in LNCaP cell line and PC-3 cell line were significant after treating with different concentrations of IL-6 for 24hs.The difference was statistically significant(P<0.05).The expression of AR,STAT3 and p38MAPK was correlated with the higher concentration of IL-6.5.IL-6 may be involved in the development of prostate cancer through AR,STAT3 and p38MAPK.6.The expression of p38MAPK was different in translation level in LNCaP and PC3 cell lines after treating with same concentrations of IL-6 for 24hs,the expression of p38MAPK in translation level in PC3 was stronger than LNCaP,IL-6 may be involved in the development of CRPC through p38MAPK.
