Objective:To investigate the effect of Rgl on the ERK signaling pathway in hepatic fibrosis induced by Pagumogonimus skrjabini infection of the liver,and to explore the possible molecular mechanism of Rgl in hepatic fibrosis.Methods:50 SD rats were randomly divided into 5 groups(n = 10).Include the normal group,the model group and the treatment groups which contains the low dose group,the middle dose group and the high dose group.Except for the normal group,every rats in each group were injected intraperitoneally with 15 cysts of Pagumogonimus skrjabini.Liver fibrosis induced by this worm was treated with low,medium and high dose Rgl solution for once a days and last for 8 weeks.The rats were sacrificeed after 8 weeks.Blood was taken and the contents of IL-1 and IL-6 were detected by ELISA.The liver tissue is divided into several parts.Some for HE staining and Masson staining to observe cell morphology and tissue fibrosis.Immunohistochemistry was used to detect the expression of P-ERK1/2 and related proteins like a-SMA,COL-1,COL-3 in liver cells that analyze the effect of Rgl on the hepatic fibrosis induced by the infection of Pagumogonimus skrjabini infection.Some are for the content of mRNA Q-PCR detection of ERK 1/2 and related pathways factors like ?-SMA,COL-1 and COL-3.Some are using Western blot to detect ERK1/2 and related pathways factors proteins like P-ERK1/2,?-SMA,COL-land COL-3.Results:Compared with the normal group,the content of IL-1 and IL-6 increased in the model group(P<0.01),and the content of IL-1 and IL-6 low dose groups decreased after the treatment of Rg1(P<0.05)while the medium dose groups and high dose groups decreased significantly(P<0.01).HE staining showed that the structure of hepatic cells was disordered,and the infiltration of fibroblasts was observed.Blue staining part was observed in Masson staining.After treatment,the protein expression of P-ERK1/2 and related factors a-SMA,COL-1,COL-3 were decreased,and the morphology of hepatocytes was improved,and the area of fiber staining was decreased.The results of Q-PCR showed that the contents of ERK1/2 stays the same result and related factors a-SMA,COL-1,COL-3 in the model group were higher than those in the normal group(P<0.01).Contrast with the model group,COL-1 treatment groups were decreased meaningfully(P<0.01).At the same time,a-SMA and COL-3 low dose groups declined(P<0.05),while the medium and high dose groups decreased significantly(P<0.01),and showed dose-dependent.The content of ERK1/2 were maintain stable but related factors like P-ERK1/2,a-SMA,COL-1,COL-3 in the model group of Western Blot band was higher than that of the normal group.Contrast with the model group,COL-1 treatment groups were decreased meaningfully(P<0.01).At the same time,?-SMA and COL-3 low dose groups declined(P<0.05),while the medium and high dose groups decreased significantly(P<0.01),and showed dose-dependent.Conclusion:Pagumogonimus skrjabini could increase the hepatic inflammation,activation the ERK signaling pathway.Then promote the transformation of HSCs to fibroblast,which lead to upregulation of COL-1 and COL-3 expression that caused by ECM accumulation which leading to liver fibrosis.Rgl may reduce the ERK signaling pathway,decrease the activation of HSCs,decrease the production of ECM,and alleviate the hepatic fibrosis caused by the infection of Pagumogonimus skrjabini.
