Objective: To detect the expression level of P62 protein in bone metastasis of lung adenocarcinoma and analyze the relationship with imaging findings, prognosis, so as to find out whether the P62 protein can be used as an indicator for prognostic evaluation of bone metastasis of lung adenocarcinoma and confirm the effect of down-regulation of P62 expression on the proliferation, invasion and metastasis of tumor cells.Methods:1?62 patients with bone metastasis of lung adenocarcinoma and 40 patients of lung adenocarcinoma without metastasistreated in the department of orthopedics of Yunnan cancer hospital from December 2007 to September 2014 were collected.Immunohistochemical method was used to detect the expression of P62 and LC3 protein in the primary tumor and bone metastasis of lung adenocarcinoma, and to analyze the difference between the two groups.2?Immunohistochemical essay was used to detect the expression of P62 and LC3?/? protein in postoperative paraffin-sections.We analyzed the relationship between the expression level of the two proteins and relevant factors such as the number of clinical bone metastases, pathological fracture, then exploring the correlation between the expression of P62 and LC3?/? protein with clinical survival time. We also made correlation analysis between the expression of the two proteins as well as their interconnections with autophagy. Multivariate analysis by SPSS 16.0 statistical analysis software was conducted to confirm whether P62 and LC3 ?/ ?protein could be used as the index of clinical prognosis(P<0.05 was considered statistically significant).Results:1? Results showed that P62 protein and LC3? protein was expressed in 62 cases of lung adenocarcinoma Paraffin section and the expression position mainly in cytoplasm;including 18 of high-expression and 22 of low-expression with regards P62 protein; 10 of high-expression and 30 of low-expression with regards to LC3?protein.Immunohistochemistry staining of 62 patients revealed the presence of P62 and LC3? protein in the cytoplasm and nucleus of lung adenocarcinoma cells,including 40 of high-expression and 23 of low- expression with regards P62 protein;23 of high-expression and 39 of low-expression with regards to LC3? protein.The expression level of P62 protein in these two different tissues was statistically significant (P=0.009).There was no significant difference in the expression level of LC3? protein in two different tissues (P=0.168).2?Soluble P62 protein in lung adenocarcinoma bone metastasis group in different gender, age, pathological fracture has no obvious difference, but different number of bone metastases in expression, P62 was increased expression in three or more than three with bone metastasis (P=0.014).The median progression free survival time was 9 months in the P62 low expression group, and the median progression freesurvival time in the high expression group was 5 months.The difference between the two groups was statistically significant by Log Rank (P=0.048).The median progression free survival time was 7 months in the LC3? low expression group, and the median progression free survival time in the high expression group was 7 months.the Log Rank test the difference between the two groups was not statistically significant (P=0.208).3?The median survival time of P62 low expression group was 12 months, and the median survival time of the high expression group was 6 months,The difference between the two groups was statistically significant by Log Rank (P=0.003).The median survival time of LC3? low expression group was 12 months,and the median survival time of the high expression group was also months, and there was no significant difference between the two groups by Log Rank test (P=0.074).4?There was no significant correlation between the expression of P62 protein and LC3? protein in Bone metastasis tissue of lung adenocarcinoma.5?4 cases of lung, lymph nodes and bone lesions in patients with surgical history of the detection of P62 protein expression in 3 tissues. The results showed that P62 protein was expressed in the primary lung tumor and lymph node metastasis, bone metastasis, and the expression of immunohistochemistry was in the order of lung lymph and bone.Conclusion:1. The expression of P62 protein in lung adenocarcinoma with bone metastases was significantly higher than that in non metastatic lung adenocarcinoma, which indicated that the role of P62 protein in metastasis was higher;2.The expression level of 2.P62 protein had no correlation with age and sex, but was positively interconnected with the number of bone metastases;3.The expression of P62 protein had negative correlation with the progression free survival of bone metastasis of lung cancer. The higher the level of P62 protein was, the shorter progression free survival might be;4.The expression of P62 protein was related to the survival time of patients with osteolytic metastasis of lung adenocarcinoma. The higher the level of P62 protein, the shorter the survival time of patients with bone metastasis of lung adenocarcinoma;5.There was no significant correlation between the expression of P62 and LC3 protein, suggesting that P62 may not regulate the bone metastasis of lung adenocarcinoma through autophagy pathway.
