Objective: To investigate the expression and clinical significance of human leukocyte antigen F-associated transcript 10 (Diubiquitin ,FAT10) and nuclear factor?Bp50 (NF-?Bp50) in infitrating ductal carcinoma of breast.Methods: This study selected the surgical resection or stinging paraffin specimens in Tumor Hospital of Yunnan Province from 2014 to 2015, including 112 cases of infitrating ductal carcinoma of breast and 53 cases of tissue adjacent to carcinoma.FAT 10 and NF-?Bp50 proteins expression in infitrating ductal carcinoma of breast and pericarcinous tissues were measured by MaxvisionTM immunohistochemistry .At the same time, the fresh infiltrating ductal carcinoma tissues and adjacent tissues were collected from 2016 to 2017, a total of 28 pairs.FAT10 and NF-KBp50 mRNA level in infitrating ductal carcinoma of breast and pericarcinous tissues were measured by Real time fluorescence quantitative PCR.The relationship between the two factors and clinicopathologic factors of infitrating ductal carcinoma of breast were analyzed from the level of protein and mRNA,and the relationship between FAT 10, NF-?Bp50 were analyzed too.Results: The positive expression rates of FAT 10 and NF- ? Bp50 protein in infitrating ductal carcinoma of breast respectively were 77.7% and 81.3% ,significantly higher than that in the pericarcinous tissues (39.6% and 45.3%), and the difference was statistically significant (P<0.001). The relative expression median of FAT 10 and NF- ? Bp50 mRNA in infitrating ductal carcinoma of breast respectively were 6.21 and 4.08,significantly higher than that in the pericarcinous tissues,and the difference was statistically significant (P<0.05). The expression of FAT10,NF- ? Bp50 protein in lymph node metastasis group or distant metastasis group were significantly higher than that in no lymph node metastasis group or distant metastasis group. The positive rates increased along with the higher histological grade and TNM stages,and the difference was statistically significant (P<0.05). But they had no statistical significance with age,tumor stage and molecular type(P>0.05). The expression of FAT10,NF- ? Bp50 mRNA were of no statistical significance with all above(P>0.05).Moreover, the protein and mRNA expression of FAT 10 were positively correlated with the expression of NF-? Bp50( r=0.62, P<0.001 and r=0.475,P=0.011).Conclusions:1. Compared to the pericarcinous tissues,the expression of FAT10,NF- ? Bp50 increased in infitrating ductal carcinoma of breast.2. The expression of FAT 10,NF- ? Bp50 protein in lymph node metastasis group or distant metastasis group were significantly higher than that in no lymph node metastasis group or distant metastasis group. The positive rates increased along with the higher histological grade and TNM stages.But they had no statistical significance with age,tumor stage and molecular type.It illustrated that the two factors were closely associated with the malignant behavior of infitrating ductal carcinoma of breast and maight participate in its progression.3. It hadn't shown statistical difference between the expression of FAT 10,NF- ? Bp50 mRNA and the clinicopathologic factors.It maight owe to the epigenetic regulation.The specific reason remained to further study.4. The expression of FAT 10 were positively correlated with the expression of NF- ?Bp50 in infitrating ductal carcinoma of breast .It predicted that the two factors may create synergies in occurrence and development of breast carcinoma.
