| Gout is a kind of dysbolismus diseases which is caused by obstacles of purine metabolism and uric acid excretion,and its clinical feature is hyperuricemia.The acute paroxysm of gout is presented by "red,swollen,hot,pain" joint,and it seriously affects the quality of patients’ life.With the improvement of living standards and the change of people’s diet,the prevalence of gout was significantly increased.Western medicine treatments of gouty arthritis are anti-inflammatory analgesic and drop hematuria acid drugs,and they are limited applications because of their serious adverse reactions and easy relapse after withdrawal.Clearing away heat and expelling pain liniment(QRQT)is for acute attack stage of gouty arthritis,aims to alleviate the symptoms of attack stage.We investigated its anti-gout effect on gouty arthritis model by sodium urate;anti-inflammatory analgesic effect by acute inflammation model,hot plate and writhing methods;the effect of arthritis factors with radioimmunoassay.1.The effect of QRQT on arthritis animals by sodium urate(1)Effect of QRQT on arthritis rats by sodium urate:Compared with the model group,QRQT(0.23g/kg),QRQT(0.46g/kg),QRQT(0.91g/kg)all could significantly decreased ankle volumes in rats,inhibit the swelling of rat joint and reduce the score of rat gait(P<0.01,P<0.05).(2)Effect of QRQT on arthritis rabbits with sodium urate:QRQT(0.46g/kg),QRQT(0.91g.kg)could significantly inhibit the swelling and the swelling rate of rat joint.Each dose group could reduce the quantity of white blood cells in rabbits’ joint cavity fluid,and the pathology score of rabbit synovial tissue(P<0.01,P<0.05),compared with the model group.2.QRQT’s anti-inflammatory effect(1)The effect of mice ear swelling by xylene with QRQT:QRQT(0.46g/kg),QRQT(0.91g.kg),QRQT(1.82g/kg)could significantly inhibit mice ear swelling by xylene(P<0.01),and enhance anti-inflammatory effects with increasing doses.(2)The effect of QRQT on rats toe edema with formalin:QRQT(0.91g/kg)at different time points after the inflammation,and QRQT(0.23 g/kg),QRQT(0,46 g/kg)at 1h,3h,4h,6h after the inflammation all could significantly inhibit rats toe edema by formaldehyde,reduce swelling and the swelling rate(P<0.01,P<0.05).3.Analgesic effect of QRQT(1)The effect of mice pain threshold with QRQT by hot plate:After 30,60,90,120 min of QRQT(1.82g/kg)administration,and after 60,90,120 min of QRQT(0.91g/kg)administration,it could significantly enhance mice pain threshold by hot plate(P<0.01,P<0.05).(2)The effect of QRQT on mice writhing by acetic acid:Three dosages of QRQT all could decrease the times of body torsion and extend the incubation period(P<0.01,P<0.05).4.Effect of inflammatory factors in arthritis rabbits by sodium urate with QRQT.IL-1β and PGE2 levels of rabbits’ synovial fluid in model group were very high.Compared with model group,each dose group of QRQT and positive control drug showed no effect on the content of IL-1β in synovial fluid,but QRQT(0.46g/kg),QRQT(0.91g.kg)could significantly inhibit the rise of PGE2 content in synovial fluid(P<0.01,P<0.05).5.The effect of serum uric acid on hyperuricemia mice with QRQTSerum uric acid was significantly increased in model group after modeling 30 min and 120 min,and it was significantly decreased in allopurinol group.QRQT(4.55 g/kg)was significantly lower the level of serum uric acid than model group after modeling 30 min,but there was no significantly difference between middle and low dose group with model group.After modeling 120 min,QRQT group was no significantly difference with model group.Conclusions:1.QRQT can inhibit the swelling rate on acute arthritis rats and rabbits by sodium urate,reduce rats’ behavioral score,and improve the pathological changes in joint synovial.It has better therapeutic effect on experimental acute gouty arthritis.2.QRQT can inhibit of mice ear edema by xylene and rats toe swelling by formaldehyde,and it has a significant analgesic effect by hot plate and acetic acid-induced pain.3.QRQT can reduce the white blood cell quantity in rabbit articular cavity fluid and prostaglandin E2(PGE2)content.In short,QRQT can the increase of capillary permeability,the oozy inflammatory cells,edema and prostaglandins levels,in order to inhibit the joint inflammation by high uric acid cause,mitigate local swelling and pain and alleviate the symptoms. |
