The "SRL.compound is a basic side came from clinical treatment of diabetes composed by the "National Medical Master" Mr.Zhou Zhongying.The compound is composed by the four herbs of Lycii Cortex,Euonymus alatus,Cpotidis Rhizoma,Corni Fructus.The compound use of the unique theory of "three hot" to set of square dialectically,which is Lycii Cortex clears dryness-heat,Cpotidis Rhizoma clears damp-heat,Euonymus alatus clears stagnant heat,Corni Fructus crue yin deficiency,and have significant clinical effect.Reports about four herbs in the prevention and treatment of diabetes and its complications are also quite a lot.This paper aims to the effective site and the active ingredient of the "SRL"compound in the treatment of diabetic vascular complications,and establish quality standards of the effective part.So we can provide experimental basis for "SRL"compound used in the clinical treatment of diabetes and its complications.This article is divided into four parts to study the effective site and the active ingredient of the "SRL" compound:Part 1:the Pharmacodynamics Comparison of the extracts of "SRL" compound in the treatment of diabetic vascular complicationsFirstly,the water extract and ethanol extract of the whole compound was prepared.Then high fat diet compound streptozotocin to establish diabetic mouse model.Taking water extract and ethanol extract to diabetic mice and determining FBG,NO,NOS,SCr and BUN,to compare the treatment efficacy of the water extract and ethanol extract in diabetic vascular complications.Determination results of the indicators show that the ethanol extract was significantly better than the water extract.Based on the above results,the extraction method of "SRL" compound was reflux extraction with 70%ethanol.Part 2:purification of effective parts of "SRL" compoundEnrich and purify the active ingredient in the "SRL" compound by macroporous resin.First,the adsorption performance as an indicator screening macroporous resin,HPD400 macroporous resin purification of compound works well.Then,using HPD400 macroporous resin to purify alcohol abstracts of "SRL" compound,distilled water,10%,30%,60%,90%ethanol as eluting solvent,UV and HPLC analysis of the active ingredient in the site of each elution.The results identified 10%-30%-60%-90%combined collection alcohol-eluting parts as the effective parts of the"SRL" compound.Part 3:the component analysis and fingerprinting of the effective parts of the"SRL" compoundFirst,UV determinate the content of the active ingredients in the effective parts of the "SRL" compound,and the total content is 80.3%;HPLC determinate the index components in the effective parts,the total composition of each index reached 23.75%.;And establish the fingerprint of the effective parts of "SRL" compound,identified the ingredient of the eight peaks in 21 common peaks,provides a comprehensive and multi-indicator method to control the quality of the effective parts of "SRL" compound.Part 4:the efficacy investigation about the effective part of "SRL" compound on treatment of diabetic vascular complicationsInvestigated the impact of the effective parts of "SRL" compound on FBQ NO,NOS,SCr,BUN,MDA,GSP of the diabetic mice.The results show that the effective parts of "SRL" compound has significant hypoglycemic effect,and can effectively regulate the level of biochemical indicators of the diabetic mice.Results show that the "SRL" compound had a therapeutic effect on diabetes and its vascular complications.
