Objective: Determination of the content of the main active substances in Compound Puerariae tablets prepared by single and mixed extraction.To elucidate the protective effect and mechanism of Compound Pueraria tablets on cardiovascular aging..Methods: Using SHIMADZU VP-ODS column(250mm×4.6,5um),The mobile phase is methanol-0.1% phosphoric acid aqueous solution,The volume flow is 0.9ml/min,The column temperature is 30 ?.The rat model of aging was established by subcutaneous injection of D-galactose,50 male Wistar rats were randomly divided into normal group,model group,Compound Pueraria tablets,high,low dose group.after 45 days later,using AU5800 automatic biochemical analyzer to mensurate the serum Hcy;using xanthine method to mensurate the serum superoxide dismutase activity in rats;using colorimetric method to mensurate glutathione peroxidase activity;using TBA method to mensurate the malondialdehyde content.Take the thoracic aorta to make pathological section.using TUNEL method to mensurate myocardial cell apoptosis rate,using JC-1 staining to mensurate the changes of mitochondrial membrane potential.using Spss16 to analyze the content of Hcy,superoxide dismutase activity,glutathione peroxidase activity,body weight,myocardial cell apoptosis rate and the percentage of JC polymer.Using HE staining the rat thoracic aorta,Histopathological changes of blood vessels were observed under light microscope,Scanning electron microscope was used to observe the microstructure of myocardium.Results: The mass concentration of puerarin and daidzein showed a good linear relationship with the peak area in the range of 15.2-236.8ug/ml and 2.5-12.5ug/ml,The average recovery rate was 98.4%,98.7%,RSD were 2.36%,3.14%,The regression equations were y = 5993.21407X-376.78985,y = 6146x+73.6200,The correlation coefficients were 0.9994,0.9993.The Hcy level,body weight were significantly different(P<0.01)between model group and the normal group,the drug group and the model group,The levels of SOD,GSH-PX and MDA in serum of rats in each group multiple comparison were significantly different(P<0.05).The apoptosis rate of myocardial cells in each group multiple comparison was significantly different(P<0.05).The percentage of JC-1 in myocardial cells of rats in each group multiple comparison were significantly different(P<0.05).In the model group,the pathological changes of vascular structure were serious,and the administration group had different degrees of improvement.Conclusion:The established method is simple,accurate and reproducible.The results provide a reliable basis for improving the quality control of compound Pueraria tablets.Dgalactose can increase the content of Hcy in the serum of aging rats.D-galactose induced aging rat model can be used to study vascular aging.Compound puerarin tablet has a good protective effect on oxidative stress in rats.Compound Gegen tablets can reduce the level of Hcy in the serum of d-galactose induced aging rats and protect the vascular structure.Compound puerarin tablet can reduce the excessive apoptosis of cardiac muscle cell.Compound puerarin tablet has the function of maintaining the stability of mitochondrial transmembrane potential of cardiac muscle cell.
