The Therapeutic Effects Of Jinbei Oral Liquid On Idiopathic Pulmonary Fibrosis In Rat Model

Objective: To observe the therapeutic effect of Jinbei oral liquid on idiopathic pulmonary fibrosis rat model and study its mechanism.Methods: Adult male SD rats were divided into six groups,including sham group,saline group(negative control group),pirfenidone group(positive control group),treraputic groups treated with different doses of Jinbei oral liquid.The model of IPF was induced by intratracheal injection of bleomycin in the model group and the treatment group,while the sham group was injected with the same volume of saline.From 7 days after operation,Jinbei groups were given by garage with different dose daily while positive control group with pirfenidone 50 mg/kg per day,and sham group and negative control group group were given same volume of saline.At twenty-eighth days after operation,the animals were sacrificed.HE and MASSON staining of lung tissues were observed under light microscope to explore pathological changes and collagen deposition condition.Concentrations of hydroxyproline(HYP),malondialdehyde(MDA)and superoxide dismutase(SOD)in lung tissues were also measured.At the same time,liver and kidney tissue were also underwent HE staining and pathological analysis.Results: Compared with the sham group,hyperemia and dark red color and uneven surface of lung tissue in saline group rats were observed with naked eyes.The lungs also had scar nodules hard texture,poor elasticity,protruding cavity.Severe alveolitis and pulmonary fibrosis could be seen under microscope after HE staining.Hydroxyproline(HYP),malondialdehyde(MDA)content significantly increased,while superoxide dismutase(SOD)activity decreased significantly in saline group rats compared with sham group ones.Compared with saline group,Jinbei oral liquid group and pirfenidone group rats had less lesions in lung than that of saline group rats.And the degree of alveolitis and pulmonary fibrosis compared with the saline group was significantly reduced.There were no obvious changes in liver and kidney tissues in all experimental rats.HYP,MDA contents in lungs of Jinbei oral liquid groups and the pirfenidone group lung tissue were significantly lower than that of saline group rats,but higher than that of sham group,while SOD activity increased significantly compared with saline group,but much lower than that of sham group.Conclusion: Jinbei oral liquid has inhibitory effect on bleomycin induced pulmonary fibrosis in rats and its mechanism might be related with regulation of oxidant/antioxidant imbalance.We believe that Jinbei oral liquid is an effective drug for treating pulmonary fibrosis,which has broad application prospect.
