Objective: To successfully replicate the model of animal pulmonary fibrosis induced by bleomycin and be discriminated with clinic idiopathic pulmonary fibrosis.Study the therapeutic effect and mechanism of Jinbei oral liquid on animal pulmonary fibrosis induced by bleomycin.Method: 1.SD rats and C57BL/6 mice pulmonary fibrosis induced by bleomycin through tracheal administration were chosen as animal models.Model evaluation was performed from death rate and cytopathic effect after four weeks.2.On the base of successful replication of the model of animal pulmonary fibrosis induced by bleomycin,the animals were treated with different doses of Jinbei oral liquid.Through animal macroscopic characterization,HE and Masson staining,Elisa detection and protein imprinting technique as well as multi-level evaluation of therapeutic effect and integrating macro and micro indicators,the therapeutic effect and mechanism of Jinbei oral liquid on animal pulmonary fibrosis induced by bleomycin was confirmed.Results:1.The optimum doses of C57BL/6 mice pulmonary fibrosis induced by bleomycin was1.5mg/kg.2.The macroscopic signs and biochemical indexes of animalmodels were combined and significant enhancement of lung index,remarkable incrassation of alveolar wall,obvious diminish of pulmonary alveoli and distinctive increase ofcollagen of the model of animal pulmonary fibrosis induced by bleomycin were confirmed.The expression levels of HYP,TGF-?,TNF-? and PDGF biomolecules in lung tissues were significantly increased detected by Elisa kit.In the lung tissue Collagen I,alpha SMA,Vimentin expression level decreased but E-cadherin expression levels rose detected by WB.Macroscopic and microscopic multiscale bleomycin induced animal pulmonary fibrosis model identification system was constructed preliminarily.3.Different doses of Jinbei oral liquid had certain therapeutic effect on the animal pulmonary fibrosis induced by bleomycin and could decrease thickness of alveolar wall,enlarge pulmonary alveoli and diminish deposition of collagen.The optimal dosage of gastric lavage of SD rats and C57BL/6 mice were 7.2 ml each per day and0.3 ml each per day,respectively.Conclusion: On the base of successful replication of the model of animal pulmonary fibrosis induced by bleomycin,macroscopic and microscopic multiscale bleomycin induced animal pulmonary fibrosis model identification system was constructed preliminarily.In bleomycin group,there was an obvious increment(p<0.01)in the content of HYP,TGF-?,TNF-? and PDGF.In addition,the protein levels of Collagen I,?-SMA and Vimentin significantly increased(p<0.01)and E cadherin levels significantly decreased(p<0.01)in bleomycin group.The groups with Jinbei oral liquid reduced HYP,TGF-?,TNF-? and PDGF content,and also depressed the levels of Collagen I,?-SMA and Vimentin,while the activity of E-cadherin were dramatically improved.Certain therapeutic effect on the animal pulmonary fibrosis induced by bleomycin by Jinbei oral liquid was confirmed and the related treatment mechanism was preliminarily discussed.
