Objective: 1.To explore the effect of EESMS on proliferation of Tca8113 cell line.2.To explore the effect of EESMS on cell cycle distribution of Tca8113 cell line.3.To explore the inducing apoptosis effect of EESMS on Tca8113 cell line.4.To explore the molecular mechanisms of EESMS by using proteomics and bioinformatics methods.Methods: MTT assay was used to evaluate the effect of EESMS on proliferation of Tca8113 cell line.The mechanism of antiproliferation activity of EESMS was investigated through morphological studies by AO/EB double staining method.Furthermore,flow cytometry was used to quantitatively detect the apoptotic rate by double labeling techniques using Annexin V FITC/propidium iodide staining analysis.Cell cycle changes were assessed using flow cytometry.The molecular mechanisms of EESMS was studied by using proteomics and bioinformatics methods.Results:1.MTT assay results showed that EESMS inhibits the proliferation of Tca8113 cells with the IC50 value of 2.085 ?g/?l.2.Analysis of AO/EB staining showed that EESMS possessed apoptosis induced antitumor activity on Tca8113 cells in a dose dependant manner.3.Flow cytometry showed that EESMS caused cell cycle arrest in G0/G1 phase.4.Annexin V FITC/propidium iodide staining analysis showed EESMS induced apoptosis of Tca8113 cells in a dose dependant manner.5.Eight proteins were identified successfully by using proteomics: Peptidyl-prolyl cis-trans isomerase A(PPIA),Receptor of activated protein C kinase 1(RACK1),L-lactate dehydrogenase B chain isoform(LDHB),Phosphoglycerate mutase 1(PGAM1),Chain A,Crystal Structure Of Human Dj-1(PARK7),Peroxiredoxin-2(PRDX2),Serpin B5(SERPIN B5),Heterogeneous nuclear ribonucleoprotein H(HNRNPH1).After analysis by biological information,those differential proteins involved in:cellular response to oxidative stress,negative regulation of gene expression,cell-cell adhesion,negative regulation of apoptotic process,oxidation-reduction process,etc.Conclusion: Our findings validated that EESMS suppresses Tca8113 cells growth and induces apoptosis throuh several biological processes.
