Osteoarthritis were the inflammatory diseases occurred in the body joint and surrounding tissue.Its clinical manifestations were the joints of redness,swelling,heat,pain and dysfunction and deformity.Glucocorticoid anti-inflammatory drugs could inhibit the agglomeration of macrophages and white blood cells in inflammation part,inhibit of phagocytosis and inhibit synthesis or release of lysosomal enzymes and the inflammatory mediators.Dexamethasone possesses better curative effect on treating osteoarthritis nowerdays,so it was chosen as the main drug in treating osteoarthritis gels.1.In this research,dexamethasone loaded in ?-cyclodextrin(?-CD)could form the micro-capsule dosage for increasing the sustained release effect.Micro-capsule was verified by UV spectrophotometric detection,sulfuric acid-fluorescence spectrum analysis and thermogravimetric analysis.Optimum preparation of micro-capsule was carried on single factor and orthogonal experiment.The drug loaded rate of microcapsule was 93.6%.The thermo-sensitive gel system was prepared by using chitosan and ?-glycerophosphate(?-GP).The single factor method was used to optimize the formulation of thermo-sensitive gel evaluated with gel time and gel temperature.Its gelation time was 5 min and its gelation temperature was 35 ?.2.Microcapsules release behavior conformed to the sustained release model in distilled water,normal saline,pH=5.8 of PBS and pH=7.4 of PBS.Dexamethasone microcapsule posssessed certain sustained release function,fitting grade 1 release equation in pharmacopoeia.Dexamethasone micro-capsule fitting degree of sustained-release dosage was 0.99,playing a preliminary slow-release effect.Then microcapsules-thermosensitive gel composite system were played dissolution experiments in distilled water,physiological saline,pH=5.8 of PBS and pH=7.4 of PBS.Dexamethasone microcapsules-thermosensitive gel composite system had a preferably sustained release,which was conformed to the level 1 release curve in the release medium.Dexamethasone microcapsules-thermosensitive gel composite system fitting degree of sustained-release dosage was 0.9,playing perfectly slow-release effect.3.Dexamethasone microcapsules-thermosensitive gel composite system possessed thermo-sensitive properties,needed to choose the appropriate sterilization method.Sterile validation of dexamethasone microcapsules-thermosensitive gel composite system had carried on the qualitative and quantitative for ultraviolet,ozone and electron beam irradiation sterilization.Electron beam irradiation sterilization can ensure sterile state of microcapsules-thermosensitive gel composite system.4.In this research,primitive cartilage cells obtaining by the pancreatic enzyme and ? collagen enzyme were used for biocompatibility evaluation of microcapsules-thermosensitive gel composite system.Biocompatibility were envaluated to cell growth for the high,medium and low doses of microcapsule-thermosensitive gel composite system,and systemic delivery and articular cavity dose of microcapsule-thermosensitive gel composite system.
