Every coin has two sides,antibiotics and other antimicrobial drugs are consu ming the living space of the human world and human wealth,which can also bring a convenient life to us.The cause of the situation is the abuse of antimicrobial drugs.The publication of World Health Organization(WHO)named?Antimicrobial Resistance: Global Report on Surveillance?reports that many factors contribute to grim situation of high level of antifungal drug resistance such as overprescribing antibiotics,which poses a threat to the health of people around the world.There is an urgent need for the development of new and effective therapeutic strategies.Photodynamic therapy(PDT)is an emerging alternative for the inactivation of fungi,it is safe and convenient.In addition,it could decrease fungal resistance potential.At present,although there were some researches that PDT can kill the Candida albicans,the effect of PDT against Candida albicans could provide a reference to the related researches.In this study,photodynamic therapy contains photosensitizer and light,toluidine blue O is a photosensitizer,the 633 nm light acts as a light source to activate toluidine blue O,then the products have an effective influence on the Candida albicans.And next step is to explore the combination effect of itraconazole and PDT or silver nanoparticles and PDT,which can provide experiences and method guide for further antifungal therapy in clinical.Main research contents and conclusions:(1)The effect of PDT was studied by colony counting method.By comparing the amount of viable cells before and after light.TBO-PDT have a significant effect on planktonic cells of Candida albicans.Photosensitized inactivation of planktonic and biofilm cultures increased with both photosensitizer concentration and irradiation time.Also,TBO-PDT can achieve the complete effect against Candida albicans.(2)The effect of PDT was studied by colony counting method.By comparing the amount of viable cells before and after light.TBO-PDT have a significant effect on biofilm of Candida albicans.Also,CLSM observations were performed to supply a better understanding of TBO-PDT in treating biofilms of Candida albicans.CLSM observations confirmed the consistence on the results of TBO-PDT on biofilm.(3)The effect of photodynamic therapy and the action of itraconazole were combined to eradicate C.albicans,and itraconazole produced significant synergistic effect on the biofilm.Moreover,its effect was also found in the planktonic cells of Candida albicans.(4)This section is about the influence of AgNPs in the TBO-PDT against Candida albicans biofilm.And the treatment is same as description above.The metabolic activity of cells in biofilm is expressed as XTT assay.And the counts of biofilm were expressed as colony-forming units(CFU/ml).There was significant change in the TBO-PDT following the incubation of AgNPs in treating C.albicans,the result shows that it is still potential to reduce the C.albicans after TBO-PDT.In conclusion,although Photodynamic Therapy is with potential prospects for direction of application in the treatment of some infections,we should promote the application through numerous available foundings.
