Expression And Significance Of MCM7、SKP2 And P27 Protein In Papillary Thyroid Carcinoma

Abstract/Summary:
Objectives To investigate the expression of MCM7 protein,SKP2 protein and P27 protein in papillary thyroid carcinoma,nodular goiter,thyroid adenoma and normal thyroid tissue,and to explore the relationship between the three proteins and the clinical and pathological features of papillary thyroid carcinoma.Explore the function of the three proteins in the occurrence and development of thyroid papillary carcinoma,try to find higher specificity and sensitivity biological indicators for the early diagnosis、treatment and prognosis of thyroid papillary carcinoma.Methods 1 In this study,a total of 140 cases of thyroid lesions were collected from June 2014 to December 2015 in the pathology department of Workers’ Hospital of Tangshan City.Among them,60 cases were papillary thyroid carcinoma,30 cases of nodular goiter,30 cases of thyroid adenoma group,20 cases of normal thyroid tissue(Selected the normal thyroid tissue outside from the adjacent thyroid papillary carcinoma for 4CM).All patients had not received preoperative radiotherapy,chemotherapy and other adjuvant therapy,and medical records information is complete.2 The expression of MCM7,SKP2 and P27 protein in thyroid papillary carcinoma group,nodular goiter group,thyroid adenoma group and normal thyroid tissue were detected by immunohistochemistry,and compared the different expressions of the three proteins in papillary thyroid carcinoma under different clinical and pathological characteristics.Explore the role of the three proteins in the occurrence and development of papillary thyroid carcinoma and the relationship between them.Results 1 The positive expression rates of MCM7 protein in papillary thyroid carcinoma group,nodular goiter group,thyroid adenoma group and normal thyroid group were 76.67%(46/60),33.33%(10/30),26.67 %(8/30)and 30%(6/20)respectively.The positive expression rate of MCM7 protein in papillary thyroid carcinoma group was significantly higher than that in thyroid nodule group,thyroid adenoma group and normal thyroid group,The difference was statistically significant(P<0.008).2 The positive expression rates of SKP2 protein in thyroid papillary carcinoma group,nodular goiter group,thyroid adenoma group and normal thyroid group were 75.00%(45/60),36.67%(11/30),30.00 %(9/30)and 10.00%(2/20)respectively.The positive expression rate of SKP2 protein in papillary thyroid carcinoma group was significantly higher than that in nodular goiter group,thyroid adenoma group and normal thyroid group,The difference was statistically significant(P<0.008).3 The positive expression rates of P27 protein in thyroid papillary carcinoma group,nodular goiter group,thyroid adenoma group and normal thyroid group were 15.00%(9/60),80.00%(24/30),76.67 %(23/30)and 90.00%(18/20)respectively.The positive expression rate of P27 protein in papillary thyroid carcinoma group was significantly lower than that in papillary thyroid group,thyroid adenoma group and normal thyroid group Statistical significance(P<0.008).4 The expression of MCM7 protein in thyroid papillary carcinoma group was not correlated with clinicopathological features such as gender,age,tumor diameter and number of lesions(P<0.05).The positive expression rate of MCM7 protein in lymph node metastasis group was significantly higher than that in non-lymph node metastasis group(P<0.05).5 The expression of SKP2 protein in papillary thyroid carcinoma group was not correlated with the clinicopathological features such as gender,age,tumor diameter and number of lesions(P>0.05).The positive expression rate of SKP2 protein in lymph node metastasis group was significantly higher than that in non-lymph node metastasis group(P<0.05).6 The expression of P27 protein in papillary thyroid carcinoma group was not correlated with gender,age,tumor diameter and number of lesions.The difference was not statistically significant(P>0.05).The positive expression rate of P27 protein in lymph node metastasis group was significantly lower than that in non-lymph node metastasis group(P<0.05).7 Correlative Analysis of MCM7、SKP2 and P27 Protein Expression in papillary thyroid carcinoma There was a negative correlation between MCM7 and P27 protein expression in papillary thyroid carcinoma(r =-0.320,P <0.05);There was a negative correlation between the expression of SKP2 and P27 protein in papillary thyroid carcinoma(r =-0.512,P<0.05);There was no correlation between SKP2 and MCM7 protein expression in thyroid papillary carcinoma(r = 0.137,P>0.05).Conclusions 1 MCM7,SKP2 and P27 proteins play an important role in the development and progression of papillary thyroid carcinoma.2 MCM7,SKP2 and P27 proteins are associated with the lymph node metastasis of thyroid papillary carcinoma.3There was a negative correlation between MCM7 and P27 protein,SKP2 and P27 protein in the occurrence of papillary thyroid carcinoma,and MCM7 had no correlation with SKP2 protein.
