Expression And Significance Of MiR-125b-1-3p In Gastric Cancer

Objective: The aim of this study is to explore the expression level of miR-125b-1-3p in gastric cancer tissues and adjacent tissues,and to analyze the correlation between the expression level of miR-125b-1-3p and clinicopathologic features.To explor its possibility as a diagnostic marker and therapeutic target in gastric cancer.Methods: The expression of miRNAs in gastric cancer tissues and corresponding adjacent tissues of 14 patients with early gastric cancer was detected by gene chip in the pre-experiment,.Tissue samples were collected from 42 patients with gastric cancer and the corresponding adjacent tissues.The total RNA was extracted from gastric cancer tissues and adjacent tissues.The expression level of miR-125b-1-3p was detected by quantitative real-time polymerase chain reaction(qRT-PCR),and to analyze the correlation between the expression level of miR-125b-1-3p and the clinicopathologic features of gastric cancer tissues.Results: The results of qRT-PCR showed that the expression of miR-125b-1-3p in gastric cancer tissues was significantly down-regulated than that in adjacent tissues.The expression level of miR-125b-1-3p in gastric cancer was only correlated with the differentiation degree,but there was no significant correlation with other clinicopathologic features such as gender,TNM stage,infiltration depth,lymph node metastasis,age and tumor size.In the gastric cancer tissues which miR-125b-1-3p was low expression,the area under the ROC curve of high / moderate differentiation degree was 0.714(P<0.05).The optimal cutoff was the relative quantity(RQ)=0.18,which the sensitivity in diagnosis of high / moderate differentiation degree was 70%,the specificity was 72.2%,the positive predictive value was 70%,and the negative predictive value was 30%.The risk of high / moderate differentiation degree in patients with RQ?0.18 is 2.33 times than that of RQ<0.18(P<0.05).It suggesting that the RQ?0.18 is a independent predictor of the gastric cancer high / moderate differentiation degree.Conclusions:The expression of miR-125b-1-3p was significantly down-regulated in gastric cancer tissues.It was found that the RQ?0.18 had a high diagnostic value in predicting gastric cancer high / moderate differentiation degree.
