Objective: MicroRNA(miRNA)is a kind of non-coding RNA.Some studies have shown that it has a similar effect to primary or tumor suppressor genes,and participates in biological events such as tumor occurrence,development,migration and apoptosis.The present study is designed to preliminarily investigate the differentially expressed microRNAs in gastric cancer tissues,and providing reference for the diagnosis and treatment of gastric cancer.Materials and Methods: 1.Three tumor tissues and paired paracancerous tissues(>5cm from tumor margin)of patients undergoing radical gastrectomy admitted in the Gastrointestinal Surgery Department of the Second Affiliated Hospital of Nanhua University between Apr.2018 and Nov.2018 were selected and the expression of miRNAs in the 3 pairs of tissues was detected by gene microarray technology.2.A total of 7 fresh gastric cancer tissues and paired paracancerous tissues(>5cm from tumor margin)removed from gastric cancer patients which were pathologically confirmed and underwent radical gastrectomy in the Gastrointestinal Surgery Department of the Second Affiliated Hospitals of Nanhua University in 2018 were collected and the accuracy of gene microarray was verified via quantitative real-time polymerase chain reaction(qRT-PCR)technique.Differentially expressed microRNAs in gastric cancer were screened and target gene prediction analysis and GO and Pathway enrichment analysis were performed for these microRNAs,and providing a theoretical basis for subsequent research.Results: 1.The results of gene microarray showed that over 1,500 miRNAs were detected,of which 17 were significantly up-regulated and 19 were significantly down-regulated.2.QRT-PCR analysis confirmed that the expressions of miR-31-3p,miR-147 b and miR-6736-3p were up-regulated(p<0.05)and that of miR-3065-5p and miR-3921 were down-regulated(p<0.05),which was consistent with the findings by gene microarray analysis.3.miR-31-3p,miR-6736-3p,miR-3065-5p,and miR-3921 have target gene intersections in TargetScan and miRDB databases,and these differentially expressed miRNAs are involved in pathological analysis of heterologous substances and chemical carcinogenesis.In the physiological process.Conclusions: 1.Expression profile of differentially expressed miRNAs in gastric cancer is obtained by gene microarray technology and miR-3921,miR-147 b,miR-3065-5p,miR-31-3p,and miR-6736-3p among others are significantly differentially expressed and possibly play important roles in the occurrence and development of this cancer.2.Bioinformatics analysis found that miR-31-3p,miR-3065-5p,miR-3921,miR-6736-3p may affect the occurrence and development of gastric cancer through various biological pathways.
