Objective: To understand the expression of SIRT1 in prostate cancer tissues,benign prostatic hyperplasia tissues and normal prostate tissues,to investigate the role of SIRT1 in the biology of prostate cancer cells,to explore the mechanism of SIRT1 in prostate cancer and to provide new targets and strategies for the diagnosis,treatment,recurrence and prognosis of prostate cancer.Methods: Immunohistochemical staining was used to detect the expression of SIRT1 in prostate cancer,benign prostatic hyperplasia and normal prostate tissues.The expression of SIRT1 in prostate cancer was also investigated by searching Cancer database ONCOMINE.The effects of SIRT1 inhibitor nicotinamide and SIRT1 siRNA on the proliferation of prostate cancer cells were studied.Finally after transfection of LNcaP cells with SIRT1 siRNA,the cell cycle and apoptosis of prostate cancer cells were detected by flow cytometry.Results: SIRT1 is highly expressed in prostate cancer tissues compared with normal prostate tissues and benign prostatic hyperplasia tissues.The expression of SIRT1 in prostate cancer is different in the tumor database ONCOMINE.SIRT1 specific inhibitor nicotinamide treatment and SIRT1 siRNA mediated knockdown of SIRT1 resulted in a marked decrease in the viability of the PCa cells.After transfection of LNcaP cells with SIRT1 siRNA,the cell cycle was inhibited and cell apoptosis was increased significantly.Conclusion: SIRT1 can promote the development,initiation and progression of prostate cancer by blocking the cell cycle and increasing the apoptosis of prostate cancer cells.SIRT1 may provide a new target and direction for the diagnosis,treatment,recurrence and prognosis of prostate cancer.
