Objective:To optimize the extraction process of polyphenols from Tetranychus citrinol by ultrasonic coupling and to study the antioxidant activity and antitumor activity of polyphenols in vitro.Methods:Using ultrasonic coupling aqueous two-phase extraction method,On the basis of single factor test,the amount of polyphenol extracted and the antioxidant activity index-iron ion equivalent was the amount of strain,The effects of ethanol concentration,ammonium sulfate concentration,ultrasonic time and ultrasonic temperature on the extraction process were studied by using the response surface design method.The extraction process of polyphenols was studied by DPPH radical,OH radical,ABTS radical scavenging ability,reducing force and other indicators to evaluate its antioxidant activity in vitro.MTT assay was used to detect the effect of polyphenols of Tetranychus angularis on the proliferation of human cervical cancer Hela cells?glioma U251 and lung cancer A549 cells.Morphological changes of cells were observed by inverted phase contrast microscope.Flow cytometry was used to analyze polyphenols Lung cancer A549 tumor cell apoptosis.Result:The optimum extraction conditions were as follows:the mass fraction of ethanol was 20.5%,the mass fraction of ammonium sulfate was 25.7%and the extraction temperature was 46?.Under these conditions,the yield of polyphenol was 59.50 mg.g-1,Equivalent to 55.62?mol.g-1,the experimental results consistent with the model predictions,and higher than the traditional impregnation extraction and ultrasonic extraction yield.The polyphenol has a strong ability to remove DPPH free radicals,ABTS free radicals and OH radicals,and has strong reducing power,which shows that it has strong antioxidant ability in vitro.The effects of polyphenols on the proliferation of human cervical cancer Hela cells?glioma U251 and lung cancer A549 cell lines were significantly inhibited by ICF at 66.82?g-mL-1?54.16 ?g·mL-1 and 49.83?g·mL-1,respectively,and showed a good dose The results showed that the polyphenols of Tetranychus angularis could induce the apoptosis of lung cancer A549 tumor cells in a dose-dependent manner,and the number of cells in each tumor cell line was significantly decreased and decreased in shape.Conclusion:The method of extraction of polyphenols from Tetranychus sinensis by ultrasonic coupling coupled aqueous method is simple,efficient and predictable.It is worthy to be popularized.The phenolic compounds of Tetraram has a significant antioxidant activity and can inhibit the proliferation of human lung cancer A549 tumor cells and induce apoptosis.
