| Polysaccharide has been suggested to have anti-tumor and immune enhacing effects recently. The aqueous extracts containing polysaccharides were extracted and purified from Trapa bispinosa Roxb,and their Physico-chemical properties were analysised in this study. The in vitro anti-tumor effect was also evaluated by MTT bioassay.An optimized extracting procedure was obtained through a L9(34)orthogonal experiment after evaluating the effects of temperature, duration of extraction,amount of water added and pH values using the extraction rate of polysaccharide as the evaluation indicator. This optimized extracting condition was finalized as temperature at 80℃, volume of water at 25 folds, duration of extraction at 4 hours and pH at 7.0 , and the cycles of extracting procedures were repeated for 3 times.The purification process for aqueous exracts includes the deproteinization and decolorization. Type II ZTC1+1 natural clarifying agent was found better than Sevege method for deproteinization, and D280 anion large pore absorbing rasin was better than H2O2. The physico-chemical properties such as the appearance, the solubility, the reaction toward iodine, the pH, and the melting point were analysised for the aqueous extracts. The melting point was more than 230℃. The infrared and ultraviolet spectrum showed a characteristic absorption of polysaccharide for Trapa bispinosa roxb aqueous extracts (TSPS). The gel filtration test revealed a mixture of compounds with very narrow distribution of molecular weigh. In addition, the protain assayes done by infraredor ultraviolet spectrum or the gel filtration test all suggested a protein binding polysaccharide in TSPS.The tests on the linearity, the recovery and the accuracy were done for the anthrone-sulfuric acid method and DNS method. The results indicated a satisfactory result when these methods were used in the assay on the Trapa bispinosa roxb polysaccharide.The in vitro anti-tumor effects of the TSPS were studied by MTT bioassay. The results showed that the inhibition by TSPS on the proliferation of the Hela cell, two cell lines of breast cancer were not significantly different from that by Cisplatin(P>0.05), while the cellular toxicity toward normal murine fibroglast cells was much greater in cisplatin than that of TSPS (P<0.01). The results of our study suggested a promising prospect in the development of aqueous extracts from Trapa bispinosa roxb as a new kind drug to cure cancer. |
