Objective:As the global epidemic of obesity and metabolic syndrome,the incidence of nonalcoholic fatty liver disease is increasing,now became the most popular liver disease exceeding viral hepatitis,but its pathogenesis has not been elucidated.Genetic factors played an important role in the process of the occurrence and development of NAFLD.Related gene mutation causes changes in the risk of NAFLD.Microsomal triglyceride transfer protein(MTTP)gene is one of the regulation of lipid metabolism genes.It has been shown that there is relationship between the gene polymorphism and hyperlipidemia,coronary heart disease,diabetes and other diseases.A large number of studies abroad have shown that there is a close relationship between MTTP gene rs1800591 polymorphism and the occurrence and development of NAFLD,but there has few research reports about the association in Han populations.Therefore,this research aimed to study the association between polymorphism rs1800591 in MTTP and non-alcoholic fatty liver disease in Han populations.Methods:To collect all the subjects' clinical indicators(gender,age,height,waist circumference,hip circumference)and laboratory indexes(alanine transaminase,aspartate transaminase,gamma glutamyl transpeptidase,uric acid,triglyceride,cholesterol ester,low density lipoprotein,high density lipoprotein).Peripheral blood DNA from 219 patients diagnosed with NAFLD and 210 control subjects was used to determine the MTTP genotype by multiple heat ligase detection reaction technology.Inter-group differences and associations were assessed statistically using t tests,Chi square and logistic analyses.Results:There were significant difference in the frequencies of rs1800591 genotype and allele between the NAFLD and control subjects(?2= 9.007,P= 0.011;?2= 9.599,P=0.002).Logistic regression analysis adjusted for confounding factors showed that the OR of carriers with the GG + GT genotype of rs1800591 developing NAFLD was 1.623,(95%CI:1.070-1.648,P=0.030),compared with those with TT genotype.The study also showed that,in contrast with T allele,there was an increased risk of NAFLD for rs1800591 G allele(OR=1.521,95%CI:1.089-2.123,P=0.014).The levels of ALT,LDL and TG were higher in GG +GT genotype than that in TT genotype,and there were significantly different among them.Conclusions:The polymorphism rs1800591 in MTTP is related to the risk of NAFLD.
