Background In recent years,Obesity,diabetes,cardiovascular and cerebrovascular diseases,fatty liver and other glylycemic metabolic disorders in the prevalence of rapid rise in China has become a major health problems.Lifestyle plays an important role in the development of glycolipid metabolic diseases.Epidemiology,authoritative guidelines and other research evidence that sedentary lifestyle and the development of glycolipid metabolic diseases are closely linked,but little in basic research reports.Objective In this study,we compared the glucose and lipid metabolism status of SD rats under different living conditions,and discussed the effect of the more comfortable conditions and difficult conditions on the metabolism of glucose and lipid in SD rats,and provided evidence for the study of glucose and lipid metabolism.Method 1.Experimental grouping: Animals maintained a week of observation in the general environment,light conditions of 13 hL/11 hD,free to eat,drinking water.According to the weight difference,the animals were randomly divided into four groups: blank control group,sedentary lifestyle group,arduous life intervention group A,hard life intervention group B.2.Experiment design: Normal group for normal light 13 hL/11 hD,free to eat ang drink for 4 weeks;sedentary lifestyle group for normal light 13 hL/11 hD,free high-sugar and high-fat diet for 12 weeks;arduous lifestyle intervention A group of light conditions for 24 hL,and placed The water environment for 4 weeks,high-fat diet for 8 weeks;arduous lifestyle intervention group B normal light 13 hL/11 hD,high-fat diet for 4 weeks,light conditions for 24 hL,and placed inthe water environment small platform in next 4 weeks,and finally continue to high fat diet for 4 weeks.3.Detection Indicator: Effects of different living conditions on sugar and lipid metabolism in rats and its mechanism: The rats were subjected to an OGTT test at 4,8,and 12 weeks,after 12 weeks,the rats were fasted for 8 hours before sacrificing.The levels of TC,TG,HDL-C,LDL-C,FBG,FINS,HOMA-IR,TNF-?,IL-6,total adiponectin and HMW adiponectin were measured at different time points.The expression of PPAR? protein and C/EBP? protein in adipose tissue were detected by Western blot.The expression of PPAR? and C/EBP? were detected by immunohistochemical staining.The expression of PPAR? and C/EBP? in adipose tissue were detected by Western blot.Adipose tissue expression and RT-PCR were used to detect the expression of PPAR? and C/EBP? in adipose tissue.Results 1.After 12 weeks,the body weight of each group was increased,the liver index of the sedentary lifestyle group was significantly higher than the normal group,while the intervention groups were compared with the sedentary lifestyle group and the liver index decreased.Normal group SD rats and the sedentary lifestyle group within 4 weeks of weight gain significantly,sedentary lifestyle group of rats hair shiny,abdominal fat deposition,gentle temperament,do not like moving,slow action.Arduous lifestyle group SD rats hair loss was obvious to the head and neck,slightly grasping the skin then the hair fall.Moreover,the animals are irritable during hard times.2.Lipid metabolism: Compared with the normal diet group,the levels of serum TG and the levels of TC,HDL-C and LDL-C were significantly different in the sedentary lifestyle group,showing the characteristics of high triglyceride lipids;the intervention group A and the sedentary lifestyle group(P> 0.05).The HDL-C level in group A was lower than that in group B(P> 0.05),and the level of HDL-C in group B was significantly lower than that in group B(P <0.05)(P <0.05).The level of LDL-C in group A and group B was significantly lower than that in group B(P <0.05).There was no significant difference between group A and group B(P> 0.05).3.Glucose metabolism: Compared with the normal group,the FBG,FINS and HOMA-IR of the sedentary lifestyle group were significantly higher than those in the normal group(P <0.05).The level of FBG,FINS and HOMA-IR in the living intervention group was significantly lower than that in the sedentary lifestyle group(P <0.05).The results of OGTT and HOMA-IR showed that the sedentary lifestyle group was higher than that of the normal group,and the living intervention group had a significant decrease(P <0.05).(P <0.05).The level of serum TNF-? in the normal group was significantly higher than that in the sedentary lifestyle group(P <0.01).4.Histopathological staining: In the sedentary lifestyle group,the pathological sections of adipose tissue and adipose tissue of adipose tissue group were few and the volume of adipose tissue was more and larger,and the life intervention group did not significantly improve the morphology of white adipose tissue.Compared with the normal group,the brown fat cells of the sedentary lifestyle group increased in volume,the adipose tissue appeared white,and the living intervention group became better in the two groups.The cell volume was significantly smaller and the number increased.5.Western blot analysis showed that PPAR?,C/EBP? protein and gene expression were down-regulated in adipose tissue of escort group,and the expression of PPAR? and C/EBP? protein in group A was significantly higher than that in healthy group The expression of PPAR? and C/EBP? in group B was higher than that in group B.The expression of PPAR? in the subcutaneous adipose tissue of the living intervention group was not significantly different from that in the cohort group.There was no significant difference between the two groups in the C/EBP? group.And there was no significant difference between the two groups.There was no significant difference in the C/EBP? between the two groups.There was no significant difference between the two groups(P> 0.05).There was no significant difference between the two groups.Group C/EBP? were significantly increased.The results of RT-PCR showed that the m RNA levels of PPAR? and C/EBP? were significantly up-regulated in group A,and there was no significant difference in life intervention group B.Conclusion The effect of sedentary lifestyle and arduous lifestyle on the metabolism of glucose and lipid in rats is different.The arduous lifestyle can effectively up-regulate the expression of PPAR?,C/EBP? in adipose tissue,improve adipogenesis differentiation,increase the secretion of adipocytokines,improve the sedentary lifestyle Lower glucose and glucose metabolism in rats.
