Objectives To observe the expression of thioredoxin(Trx)and thioredoxin-interacting protein(Txnip)in hippocampus of diabetic rats.Methods 1 Grouping and feeding 20 randomly selected weighing 290±10gSPF level of 13 week-old male GK(Goto-Kakizaki)rats as a diabetic group(DM group,random blood glucose?11.1mmol/L),and 20 healthy male Wistar rats weighing 290±10g SPF as normal control group(NC group).Standard feed,free to eat water.Laboratory should be kept clean,well-ventilated,provide the appropriate temperature,humidity and wind speed indoors,and keep plenty of light time.2 General observation Observation and record the rat's coat color,mental state,blood glucose and body weight were measured at week 2 and 10 of feeding.3 Preparation of specimens and observation indicators Two groups of rats died due to inflammation after tails were bled.After 10 weeks of feeding,10% chloral hydrate was given anesthesia(3 ml/kg),two groups of rats were killed by decapitation(DM group 17 and group 19 NC),Leave brain tissue.HE staining was used to observe the morphology of hippocampus in rats,Immunohistochemistry analysis,Western blotting(Western blot)to detect the hippocampus of rats Trx/Txnip protein content.Results 1 General observation NC rats were normal blood glucose,bright hair,good mental state,responsive and stable body weight;DM rats had a significant increase in blood glucose(After feeding for 10 weeks,fasting plasma glucose concentration NC group 5.09±1.01 vs.DM group 12.58±1.46,p<0.001),and have the performance of diabetes metabolic syndrome,apathetic,unresponsive performance.But there was no significant difference in body weight between the two groups(After 10 weeks of feeding,the NC group was 503.45±11.02 g vs.DM group 479.62±12.18 g,p>0.05,the difference was not statistically significant).2 HE staining results The hippocampal neurons in NC group were normal,the cell membrane was intact,the cytoplasm was abundant,the nucleus was intact and the cells were neatly arranged.DM group hippocampal neuronal cell morphology is irregular,the cell membrane lost a complete structure,cytoplasm and organelles decreased,cell arrangement disorder,the normal number of neuronal cells was significantly reduced.3 Immunohistochemical results The positive expression of Trx and Txnip in CA1 region of hippocampus were all brown,the expression of Trx/Txnip protein in DM group was significantly higher than that in NC group(p<0.05).4 Western blot results The expression of Trx in hippocampus of DM group was(1.94±0.27),which was significantly higher than that of NC group(1.00±0.31),the difference was statistically significant(p<0.05);The expression of Txnip in hippocampus of DM group(2.65 ±0.38)was significantly higher than that in NC group(1.00 ± 0.15),the difference was statistically significant(p <0.001).Conclusions 1 Diabetic rats hippocampal CA1 neuronal cell morphology is irregular,cell membrane loss of integrity,cytoplasm and organelles decreased,cell disorder.2Increased expression of oxidative stress-related protein Trx/Txnip in hippocampal CA1 region of diabetic rats.
