Under the current situation,the morbidity and mortality of lung cancer in the world are rising,especially in China and other developing countries [1-2].Lung cancer has become one of the most malignant tumors in the world.Lung cancer includes small cell lung cancer(SCLC)and non-small cell lung cancer(NSCLC)according to pathological type,in which non-small cell lung cancer accounts for more than 80%.Lung adenocarcinoma is the most common in non-small cell lung cancer.In recent years,the incidence of lung adenocarcinoma continues to rise and the onset age tends to be younger,in which the most are women.Lung adenocarcinoma,like other malignant tumors,is diagnosed as terminal cancer when found and losses the best period of surgery.Current,the treatment methods of lung adenocarcinoma are still dominated by surgery,chemotherapy and radiotherapy.But it does not effectively improve the overall five-year survival rate.With the development of molecular biology,targeted therapy for lung cancer has become a hot spot.The application of targeted drugs,such as Erlotinib and Gefitinib,on non-small cell lung cancer,especially lung adenocarcinoma,has made a breakthrough.In view of the discovery of targeted drugs for lung adenocarcinoma,in order to improve the survival rate of cancer,the related markers associated with the expression of lung adenocarcinoma and the indication of prognosis have become the study focus.Melanoma-associated antigen(MAGE)is just one kind of tumor specific antigens.MAGE was found from melanoma firstly,then in various malignant tumor tissues.The high expression of the antigen can drive the occurrence and development of malignant tumor,and result in poor prognosis of patients,which doesn,t express in the normal tissues.So MAGE and itsproducts can be used for tumor molecular diagnosis and immunotherapy,such as cancer vaccine.MAGE-A is one of the most important subfamily members of the MAGE family,which has a highly conserved domain and includes fifteen members,namely MAGE-A1 ~ MAGE-A15.MAGE-A protein is considered as an ideal target molecule for tumor specific antigen and tumor therapy,providing potential targets for immunotherapy of tumors.In the present study,we buy the tissue microarray associated with lung adenocarcinoma.The expression of MAGE-A family members(including MAGE-A1,-A2,-A3,-A4,-A6,-A10 and-A12)in lung adenocarcinoma,metastatic lymph node and their corresponding normal tissues was detected by immunohistochemisty in tissue microarray.Analyze its correlation with clinical biological indicators.Objective:To investigate the expressions of tumor-associated antigen(MAGE)-A family members in lung adenocarcinoma,metastatic lymph node and their corresponding normal tissues,and to explore its correlation with clinical biological indicators.Methods: The expression of MAGE-A family members in 30 cases of paracancer?lung adenocarcinoma and corresponding metastatic lymph node was detected by tissue microarray technique combined with immunohistochemistry.To explore its correlation with clinical biological indicators of patients with lung adenocarcinoma,and the difference of the expression in lung adenocarcinoma and metastatic lymph nodes.Results:1 The expression of MAGE-A in 30 cases of lung adenocarcinoma was positive in 15 cases,and the positive rate was 50%.The expression of MAGE-A in 30 cases of metastatic lymph nodes was positive in 19 cases,and the positive rate was 63.33%.The expression of MAGE-A in 30 cases of corresponding normal tissues was positive in no case,so the positive rate was0%.The results suggest that MAGE-A is a tumor associated antigen in lung adenocarcinoma.2 No correlation was found between the expression of MAGE-A familymembers protein and the age,gender,pathological grade and the number of positive lymph nodes in patients with lung adenocarcinoma(P>0.05).3 The expression of MAGE-A was also positive in metastatic lymph node,which has the positive expression in 15 cases of lung adenocarcinoma.There were 11 cases with negative expression of MAGE-A both in lung adenocarcinoma and metastatic lymph nodes.There were 4 cases with negative expression in lung adenocarcinoma,but positive in metastatic lymph nodes.There was no case which has positive expression in lung adenocarcinoma and negative result in metastatic lymph nodes.Patients,with positive expression of MAGEA in primary lung adenocarcinoma,aslo had positive expression in the metastatic lymph nodes.The expression of the protein in lung adenocarcinoma was not significantly different from metastatic lymph node(P>0.05).Conclusion: MAGE-A family,highly expressing in lung adenocarcinoma and corresponding metastatic lymph nodes,is a tumor associated antigen in lung adenocarcinoma,which can be used as a target for immunotherapy of lung adenocarcinoma.
