The volatile oil,which is one of the major components of the relieving drugs is the material basis of primary efficacy of these drugs.However,due to its volatility,oxidation and resolvability,the traditional method of spraying the volatile oil into the preparation,whether it is capsules,tablets or other common forms,there are serious problems such as the loss of effective components.The quality risk and problems of antelope cold capsules and tables was presented by the fourth quarter of 2014,the drug risk assessment conference which is held by the Hebei food and Drug Administration.Therefore,to ensure the quality of the preparation,the use of new technologies,new methods and enhancing the stability of volatile oil are important work in the development of volatile oil preparation of traditional Chinese medicine.The research will exploit the technology to improve the utilization of cyclodextrin including volatile oil,to provide a scientific basis to ehancing the stability of volatile oil of the Chinese medicine and accelerating the development of traditional Chinese medicine formulations which are contained volatile oil.Objective: To optimize the extraction of volatile oil from Forsythia suspensa and Schizonepeta tenuifolia,orthogonal experiments were used to determine the optimum extraction process.In order to ensure the stability and stability of the volatile oil components,orthogonal experiment was used to optimize the volatile oil ?-Cyclodextrin inclusion process conditions and parameters,the thermal stability of inclusion complexes was investigated,and the theoretical basis for the preparation of modern preparations for antelope cold recipe was provided.Methods:1 The optimum extraction process of volatile oil was studied by steam distillation,and the volatile oil was extracted by L9(34)orthogonal test.The effects of water addition(times),soaking time(h)extraction time(h),particle size of four factors on Forsythia,Nepeta volatile oil extraction effect,to determine the best process.2 Using three kinds of inclusion method: three methods of saturated aqueous solution,grinding method and ultrasonic method were used.By L9(34)orthogonal test,the comprehensive evaluation method was adopted,and the utilization rate of volatile oil,the rate of inclusion,the oil content of the inclusion compound aer the evaluation index,to determine the best inclusion process and method preferably.The clathrate was verified by thin layer chromatography(TLC),differential scanning calorimetry(DSC),ultraviolet spectrophotometry and microscopy.3 The volatile compounds were separated from the physical mixture at60? for seven days.The volatile oil was extracted at 1,3,5 and 7 days respectively.The volatile oil was determined by ultraviolet spectrophotometry,and the stability was compared.Results:1 The best process parameters with steam distillation method for extracting Forsythia,Nepeta mixed volatile oil: adding 10 times the amount of water,soaking for 3 hours,extraction for 4 hours,the size of the half-valve.2 The best inclusion conditions with ?-CD inclusion compound forsythia,Nepeta mixed volatile oil,mint oil: grinding method,volatile oil and ?-CD ratio of 1: 6,add water to 4 Times,the inclusion time is 45 minutes.3 In high temperature stability experiment,the inclusion of volatile oil than the physical mixture is more stable,inclusion technology can significantly improve the stability of volatile oil.Conclusions:1 Forsythia and Schizonepeta tenuifolia were extracted by orthogonal test,and the results were reasonable and reliable,which provided the basis for the extraction of volatile oil from these two flavors.2 The preferred method of inclusion in this study is simple and feasible.The inclusion rate of cyclodextrin inclusion compound prepared by this process condition is high and the stability is good,which provides experimental basis for inclusion of volatile oil in industrial production of antelope cold preparation.The study provide theoretical basis to develop modern formulations.3 The stability of volatile oil inclusion is improved,so the inclusion technology can effectively improve the stability of volatile oil for the production to provide theoretical basis.
