QJJT prescription from Sun simiao<Beiji Qianjin Yao Fang>,which is composed of rhizoma coptidis and fresh Rehmannia,play a role of heat-clearing and detoxicating,nourishing yin nourishing essence.In the clinical it was mainly used in the treatment of diabetes.After long-term clinical verification,the effect is remarkable.Modern pharmacological experimental study show that the alkaloid berberine hydrochloride in coptis chinensis can improve glucolipid metabolism and the iridoid catalpol in fresh rehmannia root can significantly improve the glucose tolerance of diabetic mice,reduce fasting blood glucose of experimental diabetes mice.Therefore,QJJT prescription were studied in my research.Research contents and results are as follows:1.Quality control study of Coptis chinensis and fresh RehmanniaAt the present,the quality of the fresh rehmannia can’t be accurately controlled.Because there only is character standard of fresh Rehmannia in the Chinese pharmacopoeia(2010 Ersion),is’t the qualitative identification and content determination method for main active ingredients.The first TLC qualitative of the main active ingredient cataptol in Fresh Rehmannia,the results of spot separation to prove that the selected methord was highly specific.At the same time using high performance liquid chromatography for the first time on Fresh Rehmannia glutinosa Libosch.’s cataptol content determination,methodological studies have shown that the methord is accurate,reliable,easy to operate,can be used for the Quality control of the Fresh Rehmannia.Determination results:three batch of fresh rehmannia from henan jiaozuo(batch number:20101125,20110409,20111115)the water content is 83.90%,84.40%,85.72%,and the content of cataptol is 7.23%、7.04%、7.38%;The content of alkaloids in rhizoma coptidis were determined using quantitative analysis of multi-components by single marker(QAMS)method in the Chinese pharmacopoeia(2010 Ersion),and the content of berberine,epiberberine and hydrastine,palmatine have a lower limit.The quality standards are better than before.Therefore,my experiment control the quality of rhizoma coptidis using this method.Determination results:rhizoma coptidis(20090704):epiberberine was 1.13%,hydrastine was 1.74%,and palmatine was 1.71%,berberine was 5.97%,higher than lower limit specified by the pharmacopoeia.2.The extract process route selection of QJJT prescriptionAccording to the physical and chemical properties and pharmacological effects of the main active ingredients,designed four extraction process routes:Gu fang preparation process,mixed reflux,Separate extract process 1(including oligosaccharides),Separate extract process 2(excluding oligosaccharides).Comparing the transfer rate of cataptol and the berberine hydrochloride in different extraction process route;At the same time,comparing the differences of different extraction process route by multicomponent chromatography;Comparing the difference between the different process routes by glucose tolerance experiment in mice.Comprehensive analysis of the experimental results,determined QJJT prescription best extraction process route:modern technology 2(flash extract fresh rehmannia,ethanol hot reflux extract rhizoma coptidis,take the extract respectively,calculate the crude drug content,according to 1:10 mixed,shake well)3.The extraction process parameters optimization of QJJT prescriptionHydrochloric berberine extraction yield and the extract rate as the evaluation index to optimize the extraction technology parameters and determine the optimal extraction process parameters by orthogonal experiment.As follows:rhizoma coptidis 12,10,10 times 70%ethanol reflux extraction three times,extracting time 1.5h,under optimum conditions hydrochloric berberine extraction rate was 90.72%,the extract rate was 20.88%;Cataptol extraction yield and the extract rate as the evaluation index to optimize the extraction technology parameters and determine the optimal extraction process parameters by star design-response surface method.As follows:fresh rehmannia 15 times 25%ethanol,extract one times,extracting time 80s,under optimum conditions cataptol extraction rate was 88.32%,the extract rate was 16.43%.Finally optimizating alcohol sinking process conditions,as follows:fresh rehmannia flash extract concentrated liquor relative density 1.10,alcohol sinking to 80%,alcohol sinking time 12h.
