Objective:To explore the roles of vitamin D and Treg/Th17 cells in the pathogenesis of AR,we detected the expression of serum vitamin D levels and the levels of Treg cells,Th17 cells in peripheral blood in AR patients.The correlation between vitamin D and Treg/Th17 cells was studied.It may provides a new clinical method for AR treatment.Methods:In our study,38 cases were selected from the outpatients of otolaryngology department of the second Hospital of Hebei Medical University who were through the relevant examination diagnosed the allergic rhinitis,as AR group or experimental group.Selected another 38 cases from the second Hospital of Hebei Medical University physical examination center,and fund the results of physical examination without related diseases,as the control group.First of all we collected the peripheral blood of the two groups subjects and each of them to prepare a common vacuum blood tube and an anticoagulant tube.Extracted about 4ml peripheral venous blood from the subjects into the vacuum blood tubes,conventionally centrifuged as required,gathered the serum of upper layer and each of them need to be placed in two tubes,one for detecting the expression level of 25 hydroxy vitamin D3 in peripheral blood using double antibodies sandwich ELISA and the calculated concentration of 25 hydroxy vitamin D3 according to OD value that had obtained.Another tube used electrochemiluminescence immunoassay(ECLI)measuring total serum vitamin D levels.At the same time,3ml peripheral blood collected from each of the subjects into the anticoagulant tube,and make the venous blood with anticoagulant mixing,measuring via flow cytometry(FCM)for CD4+CD25+Foxp3+ Treg cells and CD4+IL-17+Th17 cells expression levels,the results were showed the cell number by the percentage of CD4+CD25+Foxp3+ Treg cells in overall CD4+ Tcells and the percentage of CD4+IL-17+Th17 cells in total CD4+ T cells.All experimental data were statistically analyzed by SPSS21.0 software.Our study belongs to measurement data,and all data were testified as normal distribution via normality test data,the two groups were compared with two independent samples T-test,correlation analysis was made using Pearson correlation test,all statistics were double side probability test,?= 0.05,P<0.05 was considered statistically significant.Results:1 The total serum vitamin D level was(15.069±3.874)ng/mL in AR group and(22.346±4.260)ng/mL in control group,the total serum vitamin D level in AR group was obviously lower than that in control group(t=-7.791,P<0.05).Showed that the two group exist differences,the differences had a statistically significance.2 The 25 hydroxy vitamin D3 level was(14.139±5.013)ng/mL in AR group and(17.045±5.368)ng/mL in control group,the 25 hydroxy vitamin D3 in AR group was obviously lower than that in control group(t=-2.439,P<0.05),Showed that the content differences can be reflected in the two groups,the differences had a statistically significance.3 The percentage of CD4+CD25+Foxp3+ Treg cells in CD4+ T cells was(0.144±0.050)% in AR group and(0.560±0.142)% in control group,the percentage of CD4+CD25+Foxp3+ Treg cells in CD4+ T cells was significant lower in AR group than that in control group(t=-17.011,P<0.05).The results showed that there were some differences in the expression levels of Treg cells in the two groups,the differences had a statistically significance.4 The percentage of CD4+IL-17+Th17 cells in CD4+ T cells was(1.148±0.176)% in AR group and(0.780±0.181)% in control group,the percentage of CD4+IL-17+Th17 cells in CD4+ T cells was significant higher in AR group than that in control group(t= 8.986,P<0.05).The results showed that there were some differences in the expression levels of Th17 cells in the two groups,the differences had a statistically significance.5 Correlation analysis between the percentage of CD4+CD25+Foxp3+Tregcells in CD4+ T cells and serum 25 hydroxy vitamin D3 level was made in AR group and the result showed that there was a positive correlation between the two(r= 0.713,P<0.05).6 Correlation analysis between the percentage of CD4+IL-17+Th17 cells in CD4+ T cells and serum 25 hydroxy vitamin D3 level was made in the AR group and the result showed that there was a negative correlation between the two(r=-0.658,P<0.05).Conclusions:1 25(OH)D3 has the stable properties in the blood circulation for 2~3weeks,so it represents the level of serum vitamin D in the blood circulation.And it also been the evaluation index of vitamin D nutritional status.The total serum vitamin D level and 25-hydroxy vitamin D3 level in AR group of our test was obviously lower than that in control group,it indicated that deficiency of vitamin D was related to the incidence of AR.Vitamin D maybe becoming one of the treatments of AR,and relieving the symptoms of AR.2 The percentage of the Treg cells in CD4+ T cells was lower in AR group than that in control group,and the percentage of Th17 cells in CD4+ T cells was higher in AR group than that in control group.It is Further proved that in AR patients the number of Treg cells is decreased and the number of Th17 cells is increased.This is consistent with our knowledge of AR pathogenesis conclusion,and further validation that the Treg cells' negative immune regulation function may be suppressed.Treg cells and Th17 cells immune imbalance may be one of the important mechanisms of AR.3 Correlation analysis between the percentage of CD4+CD25+Foxp3+Treg cells in CD4+ T cells and serum 25(OH)D3 level was made in AR group and the result showed that there being a positive correlation between the two.And correlation analysis between the percentage of CD4+IL-17+Th17 cells in CD4+ T cells and serum 25(OH)D3 level was made in AR group and the result showed there being a negative correlation between the two.Illustrated that there maybe some correlation between serum vitamin D level with the imbalance of Treg/Th17 cells in AR.The lack of Vitamin D can lead to areduction in Treg cells,the increase the number of Th17 cells caused imbalance of Treg/Th17 cells,resulting in allergic rhinitis.
